【摘要】：目的：探討細(xì)胞周期轉(zhuǎn)錄因子E2F3在腎透明細(xì)胞癌以及對(duì)應(yīng)瘤旁組織中的表達(dá)情況，分析E2F3在腎透明細(xì)胞癌的發(fā)生和惡性進(jìn)展中所起的作用。 方法：通過(guò)實(shí)時(shí)定量PCR和免疫組化方法檢測(cè)E2F3在腎透明細(xì)胞癌及對(duì)應(yīng)瘤旁組織中mRNA和蛋白的相對(duì)表達(dá)情況，并分析E2F3的mRNA水平和蛋白水平表達(dá)與臨床病理資料的相關(guān)性。 結(jié)果：和對(duì)應(yīng)瘤旁組織相比，E2F3的mRNA水平和蛋白水平在腎透明細(xì)胞癌組織中顯著下調(diào)（mRNA水平0.051±0.073VS1.841±1.551，，P＜0.001；蛋白水平0.434±0.109VS0.779±0.103，P＜0.001），在TNM分期和臨床分期組間有統(tǒng)計(jì)學(xué)差異（mRNA水平0.018±0.009VS0.184±0.157，P＜0.05；蛋白水平0.407±0.115VS0.494±0.063，P＜0.05），E2F3的mRNA水平以及蛋白水平在不同年齡、性別組間無(wú)統(tǒng)計(jì)學(xué)差異（P＞0.05）。 結(jié)論：E2F3表達(dá)下調(diào)可能在腎透明細(xì)胞癌的腫瘤形成中發(fā)揮作用，且E2F3表達(dá)降低可能促進(jìn)腎透明細(xì)胞癌的惡性進(jìn)展。
[Abstract]:Aim: to investigate the expression of cell cycle transcription factor E2F3 in renal clear cell carcinoma and its adjacent tissues, and to analyze the role of E2F3 in the development and malignant progression of renal clear cell carcinoma. Methods: the relative expression of E2F3 mRNA and protein in renal clear cell carcinoma and its adjacent tissues were detected by real-time quantitative PCR and immunohistochemistry, and the correlation between the expression of E2F3 mRNA and protein and clinicopathological data was analyzed. Results: the mRNA and protein levels of E2F3 were significantly decreased in renal clear cell carcinoma (0.051 鹵0.073 vs 1.841 鹵1.551, P < 0.001; protein level 0.434 鹵0.109 vs 0.779 鹵0.103, P < 0.001), and there was a significant difference between TNM staging and clinical staging (0.018 鹵0.009 vs 0.184 鹵0.157, P < 0.05). The protein level of E2F3 was 0.407 鹵0.115 vs 0.494 鹵0.063 (P < 0. 05). There was no significant difference in the level of E2F3 mRNA and protein between different age and sex groups (P > 0. 05). Conclusion the down-regulation of the expression of E2F3 may play a role in tumor formation of renal clear cell carcinoma, and the decreased expression of E2F3 may promote the malignant progression of renal clear cell carcinoma.
