【摘要】：3維體外培養(yǎng)法(Three dimensional culture)在研究某些疾病發(fā)生發(fā)展的中存在重要意義.本研究運(yùn)用3維懸滴培養(yǎng)技術(shù),觀察前列腺良性上皮細(xì)胞系RWPE-1和BPH-1,前列腺癌細(xì)胞系PC3、LNCaP、LNCaPable和Du145在3D懸滴培養(yǎng)條件下的形態(tài)學(xué)特征.運(yùn)用實(shí)時(shí)定量RT-PCR技術(shù)比較了在普通2D培養(yǎng)與3D懸滴培養(yǎng)條件下E-鈣黏連蛋白(E-cadherin)、波形蛋白(Vimentin)、雌激素受體α與β(ERα,ERβ),G蛋白偶聯(lián)受體(GPR30)以及雄激素受體(AR)的表達(dá)變化.結(jié)果表明,與2D培養(yǎng)法相比,3D懸滴培養(yǎng)法抑制了前列腺上皮細(xì)胞發(fā)生EMT;在RWPE-1、BPH-1和PC-3細(xì)胞系中上調(diào)了ERα的表達(dá),在LNCaP與LNCaP-able中下調(diào)了AR的表達(dá).提示,與2D法相比3D懸滴培養(yǎng)法培養(yǎng)的前列腺上皮細(xì)胞具有不同的生物學(xué)特性.
[Abstract]:Three dimensional in vitro culture (three dimensional culture) is of great significance in the study of the occurrence and development of some diseases. The morphological characteristics of prostate benign epithelial cell lines RWPE-1 and BPH-1, prostate cancer cell lines PC3, LNCaP, LNCaPable and Du145 in 3D suspension culture were studied by using 3D suspension drop culture technique. The expression of E-cadherin, vimentin, estrogen receptor 偽 and 尾 (ER 偽 -ER 尾), G protein coupled receptor (GPR30) and androgen receptor (AR) in 2D culture and 3D suspension culture were compared by real-time quantitative RT-PCR. The results showed that compared with 2D culture method, 3D suspension culture inhibited EMT-producing of prostate epithelial cells, up-regulated ER 偽 expression in RWPE-1 BPH-1 and PC-3 cell lines, down-regulated AR expression in LNCaP and LNCaP-able cells. The results suggest that compared with 2D method, 3D suspension culture method has different biological characteristics.
