megsin干擾質(zhì)粒對(duì)大鼠Thy1腎炎病變的拮抗作用

【摘要】:目的應(yīng)用大鼠Thy1腎炎模型體外輸入megsin siRNA干擾質(zhì)粒,觀察下調(diào)megsin表達(dá)后對(duì)大鼠腎炎中腎小球系膜細(xì)胞和基質(zhì)增生的抑制作用。方法雄性SD大鼠共9只,隨機(jī)等分為對(duì)照組(A組),Thy1腎炎模型組(B組)和質(zhì)粒組(C組)。C組為腎炎模型加腎動(dòng)脈注射megsin siRNA質(zhì)粒,并輔以電穿孔技術(shù)促進(jìn)質(zhì)粒浸入腎臟。腎臟標(biāo)本行real-time PCR、HE染色、PAS染色和megsin、PCNA的免疫組化染色。結(jié)果與對(duì)照組比較,HE染色結(jié)果可見B組腎小球系膜細(xì)胞明顯增生伴基質(zhì)增多,而C組腎小球細(xì)胞數(shù)量明顯少于B組,但多于A組;PAS染色可見B組腎小球系膜區(qū)基質(zhì)大量增多,呈節(jié)段性分布,C組則與之相比基質(zhì)增生減少;RT-PCR和免疫組化結(jié)果顯示B組腎小球中megsin mRNA和蛋白質(zhì)水平明顯升高,與A組相比兩指標(biāo)差異均有統(tǒng)計(jì)學(xué)意義(P0.01),而在C組megsin mRNA和蛋白質(zhì)水平又明顯下降,與A、B組相比兩指標(biāo)差異有統(tǒng)計(jì)學(xué)意義(P0.05)。PCNA陽(yáng)性細(xì)胞核平均數(shù)A組為3個(gè),B組14.79個(gè),而C組僅為6.94個(gè)。統(tǒng)計(jì)顯示B組和A組、B組和C組之間差異均有統(tǒng)計(jì)學(xué)意義(P0.001)。結(jié)論注射megsin干擾質(zhì)粒以后,系膜細(xì)胞、基質(zhì)的增生均受到抑制,腎炎病變減輕,提示megsin干擾質(zhì)粒體外轉(zhuǎn)染具有拮抗大鼠Thy1腎炎的作用,這為探索以系膜細(xì)胞增生為特征的腎臟疾病的防治提供了新的思路和依據(jù)。
[Abstract]:Objective to investigate the inhibitory effect of megsin siRNA interference plasmid on Mesangial cells and matrix proliferation in rat glomerulonephritis after down-regulation of megsin expression in vitro. Methods Nine male Sprague-Dawley rats were randomly divided into two groups: control group (group A) and control group (group B) and plasmid group C (group C): nephritis model and renal artery injection of megsin siRNA plasmid, supplemented with electroporation technique to promote plasmid immersion into the kidney. The renal specimens were stained by pas and immunohistochemical staining of megsinia megsini. Results compared with the control group, the glomerular Mesangial cells in group B were obviously proliferated and matrix increased, while the number of glomerular cells in group C was significantly less than that in group B, but more in group A than in group A. The results of RT-PCR and immunohistochemistry showed that the levels of megsin mRNA and protein in glomeruli in group B were significantly higher than those in group C. Compared with group A, there were significant differences in megsin mRNA and protein levels between group A and group A (P 0.01), and there was a significant difference between group A and group A (P 0.05). The average number of megsin positive nuclei in group A was 14.79 in group A, and in group A, there were significant differences between group A and group A (P < 0.05). In group C, only 6.94. Statistical analysis showed that there were significant differences between group B and group A, group B and group C (P 0.001). Conclusion the proliferation of Mesangial cells and matrix was inhibited after injection of megsin interference plasmid, and the pathological changes of glomerulonephritis were alleviated. It suggested that megsin interference plasmid transfection in vitro could antagonize Thy1 glomerulonephritis in rats. This provides a new idea and basis for the prevention and treatment of renal diseases characterized by Mesangial cell proliferation.
