雷帕霉素對高糖誘導(dǎo)的大鼠腎系膜細(xì)胞增殖、凋亡和細(xì)胞周期的影響

【摘要】:目的:評價雷帕霉素對高糖誘導(dǎo)的腎系膜細(xì)胞增殖、凋亡和細(xì)胞周期的影響,探討其在糖尿病腎病防治中的意義。方法:體外培養(yǎng)的大鼠腎小球系膜細(xì)胞株HBZY-1分為:正常對照組、高糖組、高糖加不同濃度的雷帕霉素組,應(yīng)用CCK-8法觀察細(xì)胞增殖的變化;流式細(xì)胞術(shù)檢測各組細(xì)胞的細(xì)胞周期和凋亡情況;Real-time PCR法檢測各組細(xì)胞中血管緊張素Ⅱ(ANGⅡ)、轉(zhuǎn)化生長因子β1(TGF-β1)和血管內(nèi)皮生長因子(VEGF)的表達(dá)水平。結(jié)果:高糖誘導(dǎo)下HBZY-1的增殖水平明顯上升,凋亡水平下降,ANGⅡ,TGF-β1和VEGF的表達(dá)水平上升,而雷帕霉素具有明顯抑制作用,且有劑量依賴性,并下調(diào)ANGⅡ,TGF-β1和VEGF的表達(dá);對于細(xì)胞周期,高糖組的S期細(xì)胞明顯高于正常組(P0.05);雷帕霉素干預(yù)后,S期細(xì)胞比例減少(P0.05)。結(jié)論:雷帕霉素能夠抑制高糖狀態(tài)下HBZY-1的增殖,促進其凋亡及導(dǎo)致G1/S期阻滯,同時下調(diào)ANGⅡ,TGF-β1和VEGF的表達(dá)。
[Abstract]:Aim: to evaluate the effects of rapamycin on the proliferation, apoptosis and cell cycle of renal Mesangial cells induced by high glucose, and to explore the significance of rapamycin in the prevention and treatment of diabetic nephropathy.
