氫氧化鑭對(duì)大鼠慢性腎衰高磷血癥的改善作用研究

【摘要】:目的:研究氫氧化鑭對(duì)大鼠慢性腎衰高磷血癥的改善作用。方法:取大鼠建立慢性腎衰高磷血癥模型,再隨機(jī)分為模型組、碳酸鑭組[0.3 g/(kg·d)]、碳酸鈣組[4.2 g/(kg·d)]和氫氧化鑭高、中、低劑量組[1.5、1、0.5 g/(kg·d)],每組10只。同日上午繼續(xù)ig腺嘌呤0.2 g/(kg·d),下午ig相應(yīng)藥物,1周后停止給予腺嘌呤,連續(xù)給藥22 d,另取10只正常大鼠作為正常對(duì)照組。對(duì)各組小鼠進(jìn)行一般檢查,末次給藥后檢測(cè)大鼠腎臟系數(shù),血清中鈣、磷、甲狀旁腺素(PTH)、肌酐、尿素氮含量,以及腎臟病理學(xué)變化。結(jié)果:與正常對(duì)照組比較,模型組大鼠呈現(xiàn)慢性腎衰體征,腎臟系數(shù)和磷、PTH、肌酐、尿素氮含量均增加,鈣含量減少(P0.01),腎臟切片呈明顯病理學(xué)特征。與模型組比較,碳酸鑭組、碳酸鈣組和氫氧化鑭各劑量組大鼠慢性腎衰體征均改善,腎臟系數(shù)(除氫氧化鑭高劑量組外)和磷(除碳酸鈣組外)、PTH(除氫氧化鑭低劑量組、碳酸鈣組外)、肌酐(除氫氧化鑭低劑量組、碳酸鈣組外)、尿素氮含量均減少,鈣含量和鈣磷乘積(僅碳酸鈣組)均增加(P0.05或P0.01),其余差異無(wú)統(tǒng)計(jì)學(xué)意義;腎臟切片病理學(xué)特征好轉(zhuǎn)。結(jié)論:氫氧化鑭可改善慢性腎衰高磷血癥模型大鼠腎功能,降低血清磷含量。
[Abstract]:Objective: to study the effect of lanthanum hydroxide on hyperphosphatemia in rats with chronic renal failure. Methods: the hyperphosphatemia model of chronic renal failure was established in rats. The rats were randomly divided into three groups: model group, lanthanum carbonate group (0.3 g/(kg d), calcium carbonate group (4.2 g/(kg d) and lanthanum hydroxide high, middle and low dose groups (1.5 g/(kg / d) with 10 rats in each group. On the same day, the administration of adenine 0.2 g/(kg was continued in the morning. After 1 week of administration, adenine was given continuously for 22 days, and 10 normal rats were taken as normal control group. The kidney coefficient, serum calcium, phosphorus, parathyroid hormone (PTH), creatinine (creatinine), urea nitrogen (urea nitrogen) and pathological changes of kidney were detected in each group. Results: compared with the normal control group, the rats in the model group showed signs of chronic renal failure, renal coefficient and the contents of phosphoric acid PTH, creatinine, urea nitrogen increased, calcium content decreased P 0.01, and the renal sections showed obvious pathological characteristics. Compared with the model group, the chronic renal failure signs of rats in lanthanum carbonate group, calcium carbonate group and lanthanum hydroxide group were improved, and the renal coefficient (except lanthanum hydroxide high dose group) and phosphorus (except calcium carbonate group) and phosphorus (except lanthanum hydroxide low dose group) were improved. The contents of urea nitrogen in calcium carbonate group and calcium phosphate product (only calcium carbonate group) increased significantly (P 0.05 or P 0.01), except for lanthanum hydroxide low dose group and calcium carbonate group. The pathological features of renal sections were improved. Conclusion: lanthanum hydroxide can improve renal function and decrease serum phosphorus content in rats with chronic renal failure hyperphosphatemia.
