初生至成年大鼠膀胱ICC細(xì)胞增殖變化及其與膀胱興奮性的關(guān)系

【摘要】:目的探討初生至成年時(shí)期大鼠膀胱Cajal間質(zhì)細(xì)胞(interstitial cells of Cajal,ICC)增殖變化情況及其與相應(yīng)時(shí)間點(diǎn)膀胱興奮性的關(guān)系。方法 40只剛出生的大鼠分為2組,每組20只。第1組從出生后第1、15、30、45、60天對(duì)其膀胱進(jìn)行c-kit和Ki67染色,觀察ICC細(xì)胞的增殖情況;第2組在上述同一時(shí)間對(duì)其膀胱進(jìn)行c-kit和SM-actin染色,共聚焦顯微鏡下觀察ICC的分布密度,并測(cè)定同期膀胱肌條的興奮性。結(jié)果膀胱ICC細(xì)胞的增殖速度和分布密度從第1天開(kāi)始逐步增加,到第30天后增殖速度明顯下降,分布密度也隨之趨于穩(wěn)定。大鼠逼尿肌肌條興奮性從出生后第1天逐漸下降,第45~60天趨于穩(wěn)定。結(jié)論大鼠膀胱ICC細(xì)胞在出生后迅速增殖,到第45天分布密度趨于穩(wěn)定。膀胱興奮性隨著ICC分布密度的增加逐漸降低,并趨于穩(wěn)定。
[Abstract]:Objective to investigate the proliferation of interstitial cells of Cajal in rat bladder from birth to adulthood and its relationship with bladder excitability at the corresponding time point. Methods Forty newborn rats were divided into 2 groups, 20 rats in each group. The proliferation of ICC cells was observed by c-kit and Ki67 staining in the bladder of the first group, and the distribution of ICC was observed by confocal microscope under confocal microscope, c-kit and SM-actin staining were performed on the bladder at the same time. The excitability of bladder muscle strips at the same time was measured. Results the proliferation rate and distribution density of bladder ICC cells increased gradually from the first day to the 30th day, and the distribution density tended to be stable. The excitability of detrusor muscle strips decreased gradually from the first day after birth and stabilized on the 45th day. Conclusion Rat bladder ICC cells proliferate rapidly after birth, and the density of bladder ICC cells tends to be stable at day 45. The excitability of bladder decreased gradually with the increase of ICC density and tended to be stable.
