高糖環(huán)境下HIF1α-KIM1信號通路對糖尿病腎病腎間質(zhì)纖維化的影響

【摘要】:目的:探討高糖環(huán)境下缺氧誘導(dǎo)因子-1α(hypoxia-inducible factor-1α,HIF1α)-腎損傷分子-1(kidney injury molecule-1,KIM1)信號通路對糖尿病腎病腎間質(zhì)纖維化的影響。方法:體外培養(yǎng)人腎小管上皮細胞,分為正常對照組、高糖組、轉(zhuǎn)染對照組、HIF1α轉(zhuǎn)染組和KIM1轉(zhuǎn)染組:(1)正常對照組(5.6 mmol/L葡萄糖);(2)高糖組(30 mmol/L葡萄糖);(3)轉(zhuǎn)染對照組(30 mmol/L葡萄糖+亂序的非特異性siRNA);(4)HIF1α轉(zhuǎn)染組(30 mmol/L葡萄糖+HIF1αsiRNA);(5)KIM1轉(zhuǎn)染組(30 mmol/L葡萄糖+KIM1siRNA)。分析HIF1α、KIM1、纖維連接蛋白(fiber link protein,FN)和Ⅰ型膠原蛋白(Ⅰtype COLlagen,COL-Ⅰ)的表達。結(jié)果:高糖組的的HIF1α、KIM1、FN、COL-Ⅰ的蛋白相對表達量和mRNA相對表達量明顯高于正常對照組,差異具有統(tǒng)計學(xué)意義(P0.05)。HIF1α轉(zhuǎn)染組的HIF1α、KIM1、FN、COL-Ⅰ的蛋白相對表達量和mRNA相對表達量明顯低于轉(zhuǎn)染對照組,差異具有統(tǒng)計學(xué)意義(P0.05)。KIM1轉(zhuǎn)染組的KIM1、FN、COL-Ⅰ的蛋白相對表達量和mRNA相對表達量明顯低于轉(zhuǎn)染對照組,差異具有統(tǒng)計學(xué)意義(P0.05)。KIM1轉(zhuǎn)染組的HIF1α的蛋白相對表達量和mRNA相對表達量與轉(zhuǎn)染對照組比較并無明顯變化。結(jié)論:高糖環(huán)境下HIF1α-KIM1信號通路能夠通過調(diào)控FN、COL-Ⅰ等相關(guān)蛋白的表達影響糖尿病腎病腎間質(zhì)纖維化的發(fā)生和發(fā)展。
