香菇多糖與環(huán)磷酰胺聯(lián)合治療前列腺癌荷瘤小鼠的實(shí)驗(yàn)研究

【摘要】:目的前列腺癌具有較高的死亡率,探求其有效治療方式已成為學(xué)者的研究重點(diǎn)。本研究探討香菇多糖聯(lián)合環(huán)磷酰胺治療前列腺癌荷瘤小鼠效果及聯(lián)合治療機(jī)制,為該聯(lián)合治療方案臨床應(yīng)用提供依據(jù)。方法利用RM-1細(xì)胞建立前列腺癌皮下移植小鼠模型,將60只Balb/c荷瘤小鼠隨機(jī)平分為4組,即對(duì)照組(Control組,生理鹽水治療)、LNT組(香菇多糖治療)、CTX組(環(huán)磷酰胺治療)和LNT+CTX組(香菇多糖和環(huán)磷酰胺治療);觀察治療后荷瘤小鼠腫瘤體積比與平均存活時(shí)間,蘇木精-伊紅(HE)染色觀察治療后腫瘤組織病理學(xué)改變,檢測(cè)荷瘤小鼠脾質(zhì)量、脾指數(shù)和淋巴細(xì)胞水平確定荷瘤小鼠免疫功能情況。結(jié)果聯(lián)合治療后LNT+CTX組荷瘤小鼠腫瘤體積比達(dá)到3.51±0.80,顯著低于Control組(t=3.174)、LNT組(t=2.730)和CTX組(t=3.086),P0.05;且治療16d后,LNT+CTX組腫瘤體積比出現(xiàn)下降趨勢(shì)。HE染色結(jié)果進(jìn)一步表明,LNT組、CTX組和LNT+CTX組腫瘤組織出現(xiàn)明顯病理學(xué)改變,且LNT+CTX組病理學(xué)改變最明顯。LNT+CTX組荷瘤小鼠平均存活時(shí)間達(dá)到60d,顯著長(zhǎng)于Control組(χ~2=7.747)、LNT組(χ~2=3.653)和CTX組(χ~2=2.784),差異有統(tǒng)計(jì)學(xué)意義,P0.05。香菇多糖和環(huán)磷酰胺聯(lián)合治療后,LNT+CTX組荷瘤小鼠脾質(zhì)量[(0.28±0.08)g]、脾指數(shù)[(16.96±2.48)mg/g]及淋巴細(xì)胞[CD3+為40.67±3.13,CD4+為25.76±1.90,CD4+/CD8+為1.62±0.24]顯著低于LNT組,但高于CTX組和Control組,均P0.05。治療后各組荷瘤小鼠CD8+水平差異無(wú)統(tǒng)計(jì)學(xué)意義,P0.05。結(jié)論香菇多糖與環(huán)磷酰胺聯(lián)合作用共同減緩了腫瘤生長(zhǎng),提高了荷瘤小鼠存活時(shí)間,其可能機(jī)制是香菇多糖提高了環(huán)磷酰胺治療小鼠的免疫功能,促進(jìn)了對(duì)腫瘤殺傷。
[Abstract]:Objective Prostate cancer has a high mortality rate, and it has become the focus of scholars to explore its effective treatment.The purpose of this study was to investigate the therapeutic effect of lentinan combined with cyclophosphamide on prostate cancer bearing mice and to provide evidence for clinical application of the combined therapy.Methods RM-1 cells were used to establish subcutaneous transplantation model of prostate cancer in mice. 60 Balb/c mice were randomly divided into 4 groups: control group (control group), control group (control group), and control group (control group).The tumor volume ratio and mean survival time of tumor-bearing mice were observed after treatment with Lentinus edodes (Lentinus edodes) group (CTX group) and LNT CTX group (Lentinus edodes polysaccharides group and cyclophosphamide group).Hematoxylin-eosin (HEH) staining was used to observe the pathological changes of tumor tissues, and to determine the immune function of tumor-bearing mice by detecting the spleen weight, spleen index and lymphocyte level of tumor-bearing mice.There were obvious pathological changes in tumor tissue in CTX group.The average survival time of the tumor-bearing mice in the LNT CTX group was 60 days, which was significantly longer than that in the Control group (蠂 2 + 7.747 L NT group) and the CTX group (蠂 2 + 2.784) group (P 0.05).After treatment, there was no significant difference in CD8 level in tumor bearing mice in each group (P 0.05).Conclusion the combined action of lentinan and cyclophosphamide can slow down the growth of tumor and increase the survival time of tumor-bearing mice. The possible mechanism is that Lentinus edodes polysaccharides can improve the immune function of mice treated with cyclophosphamide and promote the killing of tumor.
