【摘要】：目的:探討SHARPIN對(duì)去勢(shì)抵抗性前列腺癌細(xì)胞LNCaP-AI壞死性凋亡關(guān)鍵因子Rip1的調(diào)控作用,以及對(duì)細(xì)胞壞死性凋亡的影響。方法:將LNCaP-AI細(xì)胞分為T(mén)NF-α+Z-VAD(caspase抑制劑)處理組與TNF-α+Z-VAD+Nec-1(Rip1抑制劑)處理組,應(yīng)用MTS檢測(cè)各組細(xì)胞的活力,研究壞死性凋亡機(jī)制在誘導(dǎo)LNCaP-AI細(xì)胞死亡中的作用。將LNCaP-AI細(xì)胞分為陰性對(duì)照組和SHARPIN干擾(si-SHARPIN)組,RT-qPCR驗(yàn)證抑制效率,通過(guò)免疫熒光等技術(shù)進(jìn)一步探討SHARPIN調(diào)控壞死性凋亡的具體分子機(jī)制。結(jié)果:與對(duì)照組相比,TNF-α+Z-VAD處理組的LNCaP-AI細(xì)胞活力下降28%(P0.05),而TNF-α+Z-VAD+Nec-1處理組的細(xì)胞在Rip1被抑制后,細(xì)胞活力無(wú)明顯改變。在LNCaP-AI細(xì)胞中,通過(guò)siRNA抑制SHARPIN表達(dá)后,Rip1表達(dá)水平上調(diào),同時(shí),LNCaP-AI細(xì)胞壞死性凋亡比例升高。結(jié)論:LNCaP-AI細(xì)胞可通過(guò)壞死性凋亡機(jī)制誘導(dǎo)死亡,下調(diào)SHARPIN可能通過(guò)激活Rip1增強(qiáng)LNCaP-AI細(xì)胞壞死性凋亡。
[Abstract]:Aim: to investigate the regulatory effect of SHARPIN on necrotizing apoptosis of LNCaP-AI cells and the effect of Rip1 on necrotic apoptosis in Ovariectomized prostate cancer cells.Methods: LNCaP-AI cells were divided into two groups: TNF- 偽 Z-VAD(caspase inhibitor (TNF- 偽 Z-VAD Nec-1(Rip1 inhibitor) and TNF- 偽 Z-VAD Nec-1(Rip1 inhibitor (TNF- 偽 Z-VAD Nec-1(Rip1 inhibitor). MTS was used to detect the cell viability and to study the role of necrotizing apoptosis in inducing the death of LNCaP-AI cells.LNCaP-AI cells were divided into negative control group and SHARPIN interference si-SHARPIN group to verify the inhibition efficiency of RT-qPCR. The specific molecular mechanism of SHARPIN regulating necrotizing apoptosis was further investigated by immunofluorescence technique.In LNCaP-AI cells, siRNA inhibited the expression of SHARPIN and up-regulated the expression of rip1, while the percentage of necrotic apoptosis in LNCaP-AI cells increased.Conclusion death can be induced by necrotic apoptosis in the cell line of 1: LNCaP-AI, and down-regulation of SHARPIN may enhance necrotizing apoptosis of LNCaP-AI cells by activating Rip1.
