【摘要】：目的：研究各種類型的尿道下裂其尿道板和包皮中的HoxA13基因轉(zhuǎn)錄及其蛋白表達,探討在尿道下裂形成過程中HoxA13的作用。 方法：按前、中、后三型分型尿道下裂,每型各10例患兒,取其包皮及尿道板,年齡3.3歲-11.6歲。取10例包莖患兒包皮作為對照,年齡3.1歲～10.4歲。取10例陰莖癌尿道作為尿道板對照,年齡48.1歲～75.6歲。免疫組織化學(xué)(IHC)和逆轉(zhuǎn)錄PCR (RT-PCR)檢測相應(yīng)組織中HoxA13的蛋白表達量和轉(zhuǎn)錄水平。 結(jié)果：RT-PCR:HoxA13轉(zhuǎn)錄水平在對照組(尿道：1.270±0.209,包皮：1.409±0.441)高于中間型(尿道板：0.684±0.228,包皮：0.848±0.338)和后型尿道下裂(尿道板：0.464±0.164,包皮：0.497±0.218)(P0.05)；前型(尿道板：1.054±0.189,包皮：1.071±0.342)轉(zhuǎn)錄水平高于后型(P0.05)。免疫組織化學(xué)：對照組(尿道13420±2636,包皮：12050±4112)HoxA13蛋白表達水平高于中間型(尿道：5238±3065,包皮：5217±1993)和后型(尿道板：2238±2217,包皮：2095±1591)(P0.05)；前型(尿道板：10450±2123,包皮：8223±3212)表達水平高于后型(P0.05)。 結(jié)論：HoxA13在尿道下裂患者包皮和尿道板組織中的表達程度可能與尿道下裂嚴重程度有關(guān)。
[Abstract]:Aim: to study the transcription and protein expression of HoxA13 gene in urethral plate and prepuce of hypospadias and to explore the role of HoxA13 in the formation of hypospadias. Methods: according to the classification of hypospadias according to the type of anterior, middle and posterior, 10 children with hypospadias were divided into three types. The prepuce and urethral plate of 10 children with prepuce and urethra, aged from 3.3 to 11.6 years, were taken as control group. The urethra of 10 cases of penile carcinoma, aged 48.1 years and 75.6 years, were used as control. The expression of HoxA13 protein and transcription level in the corresponding tissues were detected by immunohistochemistry and reverse transcription PCR RT-PCR. Results in the control group (urethral: 1.270 鹵0.209, prepuce: 1.409 鹵0.441), the transcription level of: RT-PCR: HoxA13 in the control group was higher than that in the intermediate type (urethral plate: 0.684 鹵0.228, prepuce: 0.848 鹵0.338) and posterior hypospadias (urethral plate: 0.464 鹵0.164, prepuce: 0.497 鹵0.218), and the transcription level in the former type (urethral plate: 1.054 鹵0.189, prepuce: 1.071 鹵0.342) was higher than that in the posterior type (P0.05). Chemistry: the expression levels of HoxA13 in the control group (urethral 13420 鹵2636, prepuce 12050 鹵411212) were higher than those in the intermediate type (urethra: 5238 鹵3065, prepuce: 5217 鹵1993) and the posterior type (urethral plate: 2238 鹵2217, prepuce: 2095 鹵1591p0. 05), and in the former type (urethral plate 10 450 鹵2123, prepuce: 8 223 鹵3212) were higher than those in the posterior type (urethral plate: 2238 鹵2217, prepuce: 10450 鹵2123, prepuce: 8223 鹵3212). Conclusion the expression of w HoxA13 in the prepuce and urethral plate of hypospadias may be related to the severity of hypospadias.
