微小RNA204在人腎透明細胞癌中的表達及臨床意義

【摘要】:目的研究微小RNA204(miR-204)在人腎透明細胞癌(RCCC)中的表達及臨床意義。方法收集泌尿外科行手術切除的RCCC及對應癌旁正常腎臟組織標本共65例,運用qRT-PCR技術檢測miR-204在RCCC及癌旁組織中的表達水平,統計分析miR-204表達水平與患者臨床病理資料間的相關性;采用人工合成的miR-204模擬物轉染人RCCCCaki-1細胞,分別采用qRT-PCR及Western-Blot檢測轉染后miR-204下游潛在靶點Bcl-2及SIRT1 mRNA及蛋白的表達變化。結果miR-204在RCCC組織中表達水平顯著低于對應癌旁正常腎組織(P0.05);miR-204低表達與腫瘤體積增大(3cm,P0.05)、腫瘤淋巴結轉移(P0.05)及高TNM分期(Ⅲ+Ⅳ期,P0.05)顯著相關;miR-204轉染人RCCCCaki-1細胞后可顯著降低細胞內Bcl-2及SIRT1mRNA及蛋白的表達水平(P0.05)。結論 miR-204在RCCC組織中表達下調并與腫瘤惡性臨床病理特征有關,miR-204可能通過下調Bcl-2及SIRT1的表達來抑制RCCC的發(fā)生、發(fā)展過程。
[Abstract]:Objective to study the expression and clinical significance of minimal RNA204miR-204 in human renal clear cell carcinoma (RCCC). Methods 65 cases of RCCC and adjacent normal renal tissues were collected from urological surgery. QRT-PCR technique was used to detect the expression of miR-204 in RCCC and adjacent tissues, and the correlation between miR-204 expression and clinical and pathological data of patients was analyzed statistically. Human RCCCCaki-1 cells were transfected with synthetic miR-204 mimics. QRT-PCR and Western-Blot were used to detect the expression of Bcl-2, SIRT1 mRNA and protein downstream of miR-204. Results the expression of miR-204 in RCCC tissues was significantly lower than that in normal renal tissues adjacent to cancer.
