腺病毒介導(dǎo)的PDCD4基因過表達促進腎癌BTG1蛋白的表達和786-O細胞的凋亡

【摘要】:目的探討過表達抑癌基因PDCD4的腺病毒對腎透明細胞腺癌細胞株(786-O)的影響及腎臟注射后對腎癌的治療效果。方法體外構(gòu)建過表達抑癌基因PDCD4的腺病毒;培養(yǎng)786-O細胞株,轉(zhuǎn)染腺病毒體外干預(yù),觀察細胞的死亡情況;將SD大鼠隨機分為對照組和移植組,兩組均在前肢腋部皮下注射786-O細胞株構(gòu)建腎癌模型,造模14 d后,對照組瘤中注射0.1 ml D-Hanks,移植組則注射含1×10~(11)vp/m L的表達抑癌基因PDCD4的腺病毒的D-Hanks 0.1 ml。結(jié)果體外觀察轉(zhuǎn)染病毒后,786-O細胞的生長活力下降,死亡率升高;體內(nèi)注射病毒后,可以觀察到腎臟中抑癌基因PDCD4的上調(diào)和BTG1蛋白表達水平的升高。結(jié)論過表達抑癌基因PDCD4的腺病毒注射治療腎癌具有可行性。
[Abstract]:Objective to investigate the effect of adenovirus overexpression of tumor suppressor gene PDCD4 on renal clear cell adenocarcinoma cell line 786-O). Methods Adenoviruses expressing tumor suppressor gene PDCD4 were constructed in vitro. 786-O cell line was cultured and transfected with adenovirus in vitro to observe the cell death. SD rats were randomly divided into two groups: control group and transplantation group. Both groups were injected with 786-O cell line subcutaneously into the armpit of forelimb to construct renal cell carcinoma model, and the model was made 14 days later. In the control group, 0.1 ml D-Hanks were injected into the tumor. The transplanted group was injected with D-Hanks 0.1 ml of adenovirus containing 1 脳 10 ~ (-1) v / m L PDCD4. Results after transfection of the virus in vitro, the expression of tumor suppressor gene PDCD4 was observed. The growth activity of 786-O cells decreased and the mortality increased. After the virus was injected into the body. The upregulation of tumor suppressor gene PDCD4 and the increase of BTG1 protein expression can be observed in kidney. Conclusion it is feasible to treat renal cell carcinoma with adenovirus injection with overexpression of tumor suppressor gene PDCD4.
