丙泊酚對小鼠小腦皮層分子層和顆粒細(xì)胞層感覺信息傳遞的影響

  [目的]丙泊酚是一種中樞神經(jīng)系統(tǒng)抑制劑,在體內(nèi)條件下劑量依賴性的抑制大腦新皮層的信息處理。然而,丙泊酚對活體動物小腦皮質(zhì)分子層感覺信息處理的影響還不清楚。因此,我們在烏拉坦麻醉麻醉下,應(yīng)用電生理學(xué)和藥理學(xué)方法研究丙泊酚對感覺刺激誘發(fā)小鼠小腦分子層場電位反應(yīng)的影響。[方法]6-8周齡的成年ICR小鼠通過腹腔注射體重為1.3g/kg烏拉坦麻醉后,進(jìn)行氣管插管以避免呼吸道堵塞,然后將小鼠固定在定制的立體定位裝置上。在小腦相應(yīng)部位作一個防水外流灌流槽后,在小腦Crus Ⅱ區(qū)的相對應(yīng)部位行開顱手術(shù),鉆一個直徑為1-1.5mm的小孔,暴露記錄部位的小腦表面,并用蠕動泵在暴露腦表面持續(xù)灌流含氧的人工腦脊液(artificial cerebrospinal fluid, ACSF)o使用Axopatch-200B膜片鉗放大器(Molecular Devices, Foster City, CA)記錄感覺刺激誘發(fā)小腦皮層分子層場電位,并通過D/A轉(zhuǎn)換器1440和Clampex 10.3軟件獲取感覺刺激誘發(fā)場電位活動的數(shù)據(jù)。記錄電極內(nèi)填充的是人工腦脊液,阻抗在3-5 MΩ之間。丙泊酚和gabazine購置...
