丙泊酚對(duì)小鼠小腦浦肯野細(xì)胞活動(dòng)及感覺(jué)信息傳遞的影響

  丙泊酚對(duì)小腦皮層浦肯野細(xì)胞自發(fā)性活動(dòng)的影響[目的]丙泊酚,2,6-雙(1-甲基乙基)苯酚乳劑,是一種快速靜脈鎮(zhèn)靜催眠藥,廣泛用于麻醉的誘導(dǎo)與維持,也用于特護(hù)病房(ICU)患者的鎮(zhèn)靜。丙泊酚對(duì)整個(gè)中樞神經(jīng)系統(tǒng)均有抑制作用,其最常見(jiàn)副作用是劑量依存地抑制心肺功能。已證實(shí)丙泊酚具有活化γ氨基丁酸(GABA)A受體、抑制N-甲基-D-天門冬氨酸(NMDA)受體,并通過(guò)慢速鈣離子通道調(diào)節(jié)鈣內(nèi)流。在離體條件下,丙泊酚可通過(guò)活化GABAA和甘氨酸受體對(duì)脊髓、海馬和下丘腦神經(jīng)元的活動(dòng)產(chǎn)生抑制作用,然而,丙泊酚對(duì)活體動(dòng)物小腦皮層神經(jīng)元活動(dòng)的影響尚不清楚。因此,我們應(yīng)用細(xì)胞貼附式記錄和藥理學(xué)手段研究丙泊酚對(duì)烏拉坦麻醉小鼠浦肯野細(xì)胞自發(fā)性活動(dòng)的影響,旨在闡明靜脈麻醉藥丙泊酚對(duì)小腦皮層浦肯野細(xì)胞自發(fā)性單純性放電和復(fù)雜性放電的影響機(jī)制。[方法]成年ICR小鼠(6-8周齡)通過(guò)腹腔注射烏拉坦麻醉(1.3g/kg體重)后,行氣管插管以保證呼吸道暢通,然后將動(dòng)物固定在自制的腦立體定位儀上。在小腦部位制作一個(gè)灌流槽后,在小腦CrusⅡ區(qū)相對(duì)應(yīng)處行開(kāi)顱手術(shù),鉆一個(gè)直徑約為1-1.5mm小孔,暴露記錄部位的小腦表面,輕輕去...
