NMDA受體在小鼠小腦皮層浦肯野細(xì)胞自發(fā)性活動中的作用機制

[目的]N-甲基-D-天冬氨酸受體是離子型谷氨酸受體,它參與神經(jīng)元突觸后反應(yīng)以及調(diào)控突觸前神經(jīng)遞質(zhì)的釋放。在中樞神經(jīng)系統(tǒng)中,它對突觸傳遞的調(diào)控起著關(guān)鍵作用。眾所周知,NMDA受體在哺乳動物的小腦皮層、顆粒細(xì)胞膜、平行纖維以及分子層中間神經(jīng)元上廣泛表達。在離體條件下,NMDA受體的活化增強浦肯野細(xì)胞的抑制性突觸后電流。然而,在在體條件下,NMDA對小腦神經(jīng)元的活動影響還不清楚。因此,在我們的實驗中,應(yīng)用電生理膜片鉗記錄和藥理學(xué)方法,探索外源性NMDA對烏拉坦麻醉小鼠小腦皮層浦肯野細(xì)胞自發(fā)性活動的影響,了解NMDA對小鼠小腦皮層浦肯野細(xì)胞自發(fā)性簡單峰電位放電的影響機制。[方法]實驗選取25只6-8周齡成年ICR小鼠,并對其進行腹腔注射烏拉坦麻醉(1.3 g/kg)。待40分鐘麻醉結(jié)束后,小鼠行氣管插管術(shù),使其呼吸道保持通暢,并將小鼠固定在腦立體定位裝置上。在小腦部位制作一個圓形的灌流槽,對小鼠小腦蚓部行開顱手術(shù),打孔直徑約為1-1.5 mm,暴露小腦表面的記錄區(qū)域,并輕輕地揭掉硬腦膜,使用蠕動泵在小腦表面持續(xù)不斷地灌流(0.4 ml/min)含95%02和5% CO2的人工腦脊液。用體溫維...
