【摘要】：目的:測(cè)定成年雄性SD大鼠經(jīng)不同濃度七氟烷麻醉不同時(shí)間后血中游離六氟異丙醇(HFIP)的濃度,并分析游離HFIP含量與七氟烷麻醉濃度、麻醉時(shí)間之間的關(guān)系。方法:取健康成年雄性SD大鼠100只,隨機(jī)分為四組,每組25只,分別接受0.5MAC、1MAC、1.5MAC、2MAC濃度的七氟烷麻醉。七氟烷麻醉氣體由麻醉機(jī)上的揮發(fā)罐輸向大鼠專用麻醉裝置。麻醉機(jī)輸出七氟烷的實(shí)際濃度經(jīng)氣相色譜儀進(jìn)行校準(zhǔn)。各組大鼠分別麻醉0.5h、1h、2h、4h、6h后取其動(dòng)脈血7毫升,用二次平衡法測(cè)定血中游離六氟異丙醇濃度。求出每組數(shù)據(jù)平均值,分別繪制“吸入七氟烷濃度-血中游離六氟異丙醇濃度”關(guān)系曲線及“麻醉時(shí)間-血中游離六氟異丙醇濃度”關(guān)系曲線,分析HFIP含量與七氟烷濃度、麻醉時(shí)間之間的關(guān)系。結(jié)果:各組SD大鼠經(jīng)七氟烷麻醉后血中均能檢測(cè)出游離HFIP。組內(nèi)比較:各組大鼠血中游離HFIP濃度隨時(shí)間變化趨勢(shì)基本一致,即0.5h至4h間HFIP濃度隨麻醉時(shí)間延長(zhǎng)而逐漸增加,HFIP峰值出現(xiàn)在接受七氟烷麻醉后第四小時(shí),在第4至第6小時(shí)間呈下降趨勢(shì);組間比較:0.5MAC組各時(shí)點(diǎn)游離HFIP濃度為四組中最低者;而1.0MAC和1.5MAC組各時(shí)點(diǎn)游離HFIP濃度均較高,且都明顯高于2.0MAC組,但1.5MAC組血中游離HFIP隨麻醉時(shí)間變化趨勢(shì)最為平穩(wěn)。結(jié)論:血中游離HFIP濃度隨七氟烷麻醉時(shí)間的延長(zhǎng)呈現(xiàn)先增高而后降低的趨勢(shì);中等濃度七氟烷麻醉組血中游離HFIP濃度水平高于低濃度麻醉組,但繼續(xù)增大七氟烷麻醉濃度(2.0MAC),血中所產(chǎn)生的游離HFIP濃度反而降低。
[Abstract]:Aim: to determine the concentration of free hexafluoropropanol (HFIP) in blood of adult male SD rats after sevoflurane anesthesia with different concentrations of sevoflurane, and to analyze the relationship between free HFIP content and sevoflurane anesthetic concentration and anesthetic time. Methods: 100 healthy adult male SD rats were randomly divided into four groups (25 rats in each group). Sevoflurane anesthetic gas is transported from volatile tank on anaesthesia machine to special anesthetic device in rats. The actual concentration of sevoflurane from anaesthesia machine was calibrated by gas chromatograph. Rats in each group were anesthetized for 0. 5 h, 1 h, 2 h and 4 h for 6 h. After 6 hours, 7 ml of arterial blood was taken, and the concentration of free hexafluoropropanol in blood was determined by two times equilibrium method. The average values of each group were calculated, and the curves of "inhaled sevoflurane concentration-blood free hexafluoropropanol concentration" and "anesthetic time-blood free hexafluoropropanol concentration" were drawn, respectively. The HFIP content and sevoflurane concentration were analyzed. The relationship between anesthetic time. Results: free HFIP. could be detected in the blood of SD rats after sevoflurane anesthesia. Comparison within the group: the concentration of free HFIP in blood of rats in each group showed the same trend with time, that is, the concentration of HFIP increased gradually with the prolongation of anesthesia time between 0.5 h and 4 h, and the peak value of HFIP appeared in the fourth hour after sevoflurane anesthesia. From the 4th to the 6th, there was a downward trend. Comparison between groups: free HFIP concentration in 0.5MAC group was the lowest in the four groups at each time point. The concentration of free HFIP in 1.0MAC and 1.5MAC groups was higher than that in 2.0MAC group at each time point, but the trend of free HFIP in 1.5MAC group was the most stable with anesthesia time. Conclusion: with the prolongation of sevoflurane anesthesia time, the concentration of free HFIP in blood increased first and then decreased. The level of free HFIP in the middle concentration sevoflurane anesthesia group was higher than that in the low concentration anesthesia group, but it continued to increase the sevoflurane anesthetic concentration (2.0MAC), but the free HFIP concentration in the blood decreased.
