【摘要】：目的研究肝細(xì)胞生長因子(HGF)在先天性肥厚性幽門狹窄(CHPS)大鼠幽門的表達(dá)情況,探討HGF在CHPS起病中的作用。方法將受孕后的SD大鼠隨機(jī)分為模型組與對照組,自孕期第2天始至新生鼠生后21 d,模型組經(jīng)尾靜脈注射給予L-NAME,對照組經(jīng)相同途徑給予同等劑量的生理鹽水。新生鼠生后第1、7、14、21天分別稱重,生后第21天麻醉處死,留取幽門組織,HE染色后顯微鏡下測量幽門肌層厚度。RT-PCR半定量幽門組織的HGF mRNA的表達(dá)。結(jié)果與對照組新生鼠比較,模型組新生鼠體重增長明顯緩慢,體重較低,兩組間差異有統(tǒng)計(jì)學(xué)意義(P0.01)。與對照組新生鼠比較,模型組幽門肌明顯肥厚,差異有統(tǒng)計(jì)學(xué)意義(P0.01)。與對照組新生鼠比較,模型組新生鼠HGF mRNA的表達(dá)升高,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論幽門組織中HGF的表達(dá)增加可能與CHPS的發(fā)病有關(guān)。
[Abstract]:Objective to investigate the expression of hepatocyte growth factor (HGF) in (CHPS) rats with congenital hypertrophic pyloric stenosis and the role of HGF in the pathogenesis of CHPS. Methods SD rats were randomly divided into model group and control group. From the second day of pregnancy to 21 days after birth, the model group was injected with L-NAME via tail vein, and the control group was given the same dose of normal saline in the same way. The neonatal rats were weighed on the 1st day after birth and were anesthetized and killed on the 21st day after birth. The thickness of the myometrium of the pylorus and the expression of HGF mRNA in the RT-PCR semi-quantitative pyloric tissue were measured under the microscope after HE staining. Results compared with the control group, the body weight of the newborn rats in the model group was significantly slower and lower than that in the control group (P0.01). Compared with the control group, the model group showed significant hypertrophy of pyloric muscle (P0.01). Compared with the control group, the expression of HGF mRNA in the model group was significantly higher than that in the control group (P0.05). Conclusion the increased expression of HGF in pyloric tissues may be related to the pathogenesis of CHPS.
