邵氏“五針法”對哮喘模型大鼠a-SMA及FN表達影響的實驗研究

【摘要】:目的 本課題通過針刺哮喘模型大鼠“肺俞”、“大椎”、“風門”穴,觀察邵氏“五針法”對各組哮喘模型大鼠肺組織和血清中平滑肌肌動蛋白-a(a-SMA)及纖維連接蛋白(FN)表達的影響以及對肺組織病理變化的影響,探討邵氏“五針法”治療哮喘的機理及其對哮喘模型大鼠氣道重塑的作用機制,為針灸治療哮喘提供科學的實驗依據(jù)。 方法 選取健康雄性清潔級SD大鼠40只(體重140±20g),隨機分成空白對照組、模型對照組、藥物治療組和針刺治療組,每組10只。采用腹腔注射卵蛋白(OVA)致敏,霧化吸入1%的OVA溶液(每日1次,共7次)激發(fā)建立哮喘大鼠模型。針刺治療組于每日激發(fā)后針刺哮喘大鼠“肺俞”(雙)、“大椎”、“風門”(雙)穴治療;藥物治療組于每日激發(fā)后腹腔注射0.05%地塞米松磷酸鈉溶液治療;模型對照組激發(fā)后不做特殊處理;空白對照組腹腔注射及霧化吸入時,用生理鹽水代替卵蛋白進行操作。末次治療后6h內(nèi)水合氯醛麻醉大鼠,取眼球血后脫頸椎處死大鼠,取右中下肺組織,石蠟包埋,制備病理切片,行蘇木精-伊紅(HE)染色及a-SMA、FN肺組織免疫組化染色和血清ELISA檢測,觀察大鼠肺組織形態(tài)學改變以及a-SMA、FN在哮喘模型大鼠肺組織和血清中的表達。 結(jié)果 1.各組大鼠行為體征觀察 模型對照組大鼠與空白對照組比較,出現(xiàn)噴嚏,嗆咳,流涕,喘息,呼吸急促,精神不振,反應遲鈍;空白對照組大鼠行動敏捷,未見異常;藥物治療組、針刺治療組大鼠噴嚏,嗆咳,喘息等癥狀較模型對照組明顯減輕,反應靈敏。 2.各組大鼠肺組織形態(tài)學觀察 模型對照組大鼠氣道黏膜上皮受損脫落,黏膜下層和平滑肌層增生、變形;氣道壁增厚,管腔變窄,管腔內(nèi)大量分泌物;肺間質(zhì)大量炎癥細胞浸潤,肺泡壁破裂變形,肺泡間隔結(jié)構(gòu)紊亂;血管增生,血管管壁增厚。藥物治療組、針刺治療組氣道結(jié)構(gòu)基本完整,肺間質(zhì)炎癥細胞浸潤較少,肺組織結(jié)構(gòu)基本正常?瞻讓φ战M大鼠氣道結(jié)構(gòu)完整,無明顯脫落和變形,肺間質(zhì)未見炎癥細胞浸潤,肺組織形態(tài)結(jié)構(gòu)正常。 3.各組大鼠肺組織a-SMA的比較 空白對照組大鼠肺組織中少見胞漿呈棕黃色染色的a-SMA陽性細胞;模型對照組大鼠肺組織內(nèi)陽性表達明顯增多,與空白對照組比較,差異具有統(tǒng)計學意義(P<0.01);針刺治療組及藥物治療組與模型對照組比較,肺組織內(nèi)陽性表達明顯減少,差異具有統(tǒng)計學意義(P<0.01),但仍高于空白對照組;針刺治療組與藥物治療組比較,差異具有統(tǒng)計學意義(P<0.01)。 4.各組大鼠肺組織FN的比較 空白對照組大鼠肺組織中少見胞漿呈棕黃色染色的FN陽性細胞;模型對照組大鼠肺組織內(nèi)陽性表達明顯增多,與空白對照組比較,,差異具有統(tǒng)計學意義(P<0.01);針刺治療組及藥物治療組與模型對照組比較,肺組織內(nèi)陽性表達明顯減少,差異具有統(tǒng)計學意義(P<0.01),但仍高于空白對照組;針刺治療組與藥物治療組比較,差異具有統(tǒng)計學意義(P<0.01)。 5.各組大鼠血清a-SMA的比較 模型對照組與空白對照組相比,大鼠血清中a-SMA濃度顯著增高,差異具有統(tǒng)計學意義(P<0.01);針刺治療組及藥物治療組與模型對照組比較,a-SMA濃度明顯降低,差異具有統(tǒng)計學意義(P<0.01),但仍高于空白對照組;針刺治療組與藥物治療組比較,差異具有統(tǒng)計學意義(P<0.01)。 6.各組大鼠血清FN的比較 模型對照組與空白對照組相比,大鼠血清中FN濃度顯著增高,差異具有統(tǒng)計學意義(P<0.01);針刺治療組及藥物治療組與模型對照組比較,F(xiàn)N濃度明顯降低,差異具有統(tǒng)計學意義(P<0.01),但仍高于空白對照組;針刺治療組與藥物治療組比較,差異具有統(tǒng)計學意義(P<0.01)。 結(jié)論 1.邵氏“五針法”能夠有效改善哮喘模型大鼠的癥狀和行為體征。 2.邵氏“五針法”能夠有效改善哮喘模型大鼠的肺組織形態(tài)學改變。 3.邵氏“五針法”能夠降低哮喘模型大鼠肺組織中a-SMA和FN的陽性表達。 4.邵氏“五針法”能夠降低哮喘模型大鼠血清中a-SMA和FN的陽性表達,且二者在各組哮喘模型大鼠肺組織和血清中的表達具有一致性,并呈正相關(guān)。 邵氏“五針法”能明顯降低哮喘模型大鼠肺組織和血清中a-SMA及FN的陽性表達,改善氣道重塑,說明降低a-SMA、FN的陽性表達是邵氏“五針法”治療哮喘的作用途徑之一。
