【摘要】：目的基于內(nèi)毒素脂多糖(LPS)模型,探討六味五靈片對何首烏致大鼠特異質(zhì)肝損傷的防治作用。方法將80只SD大鼠隨機分為對照組,LPS(2.8 mgNg)組,單獨何首烏(生藥2.16 gNg)組,LPS(2.8 mg/kg)+何首烏(生藥2.16 g/kg)組,LPS(2.8 mg/kg)+何首烏(生藥2.16 gNg)+六味五靈片低、中、高劑量(0.4、0.8、1.6 g/kg)組及LPS(2.8 mg/kg)+何首烏(生藥2.16g/kg)+陽性藥聯(lián)苯雙酯(2mg/kg)組。按組別分別ig給予相應劑量的六味五靈片和聯(lián)苯雙酯,每日1次,連續(xù)給藥2d,第3天除對照組和LPS組ig等量蒸餾水外,按組別分別ig給予何首烏醇提物,3 h后按組別尾ivLPS,給予LPS7 h后戊巴比妥鈉麻醉大鼠,下腔靜脈取血并采集肝組織標本。比色法檢測血漿中丙氨酸轉(zhuǎn)氨酶(ALT)、天冬氨酸轉(zhuǎn)氨酶(AST),HE染色觀察肝組織病理學變化,TUNEL檢測分析肝細胞凋亡,ELISA法檢測血漿中腫瘤壞死因子-α(TNF-α)、白細胞介素-1β(IL-1β)、白細胞介素-6(IL-6)、γ干擾素(IFN-γ)水平,免疫組化染色觀察肝組織核轉(zhuǎn)錄因子-κB(NF-κB)p65的表達。結(jié)果六味五靈片能明顯降低何首烏誘導的特異質(zhì)肝損傷大鼠血漿中ALT和AST水平(P0.05),減輕肝組織病理損傷和肝細胞凋亡,明顯抑制NF-κB p65的表達(P0.05),同時明顯降低血漿中TNF-α等炎癥因子水平(P0.05、0.01)。結(jié)論六味五靈片可通過抑制NF-κB信號通路,減輕炎癥反應,防治何首烏誘發(fā)的特異質(zhì)肝損傷。
[Abstract]:Objective to investigate the preventive and therapeutic effects of lipopolysaccharide (LPS) (LPS) on liver injury induced by Polygonum multiflorum in rats. Methods 80 SD rats were randomly divided into 2. 8 mgNg control group and 2. 8 mg/kg (2. 8 mg/kg) and 2. 16 g/kg (crude drug 2. 16 gNg) groups. The high dose group (0.4 mg/kg 0.81.6 g/kg) and the LPS (2.8 mg/kg) Polygonum multiflorum (2.16g/kg) positive drug biphenyl diester (2mg/kg) group. According to group ig, the corresponding doses of Liuwei Wuling tablets and biphenyl diesters were given once a day for 2 days. On the third day, the same amount of distilled water was given to the control group and the LPS group, except for the same amount of distilled water as ig in the control group and the LPS group. The alcohol extract of Polygonum multiflorum was given by ig for 3 h, then the rats were anesthetized with pentobarbital sodium after LPS7 for 3 h. The blood was collected from the inferior vena cava and the liver tissue was collected. Detection of alanine aminotransferase (ALT), aspartate transaminase (AST) in plasma by colorimetric assay and detection of hepatic histopathology by HE staining; Tunel assay; apoptosis of hepatocytes; Elisa method for detection of plasma tumor necrosis factor- 偽 (TNF- 偽), interleukin-1 尾 (IL-1 尾), interleukin (IL) The levels of 6 (IL-6), interferon 緯 (IFN- 緯), The expression of nuclear factor-魏 B (NF- 魏 B) p65 was observed by immunohistochemical staining. Results Liuwei Wuling tablet could significantly reduce the levels of ALT and AST in plasma of rats with liver injury induced by Polygonum multiflorum (P0.05), alleviate the pathological injury of liver tissue and the apoptosis of hepatocytes. The expression of NF- 魏 B p65 was significantly inhibited (P0.05), and the level of inflammatory cytokines such as TNF- 偽 in plasma was significantly decreased (P0. 05 鹵0. 01). Conclusion Liuwei Wuling tablet can attenuate the inflammatory reaction by inhibiting NF- 魏 B signaling pathway, and prevent and treat the liver injury induced by Polygonum multiflorum.
