樺褐孔菌對實驗性Ⅱ型糖尿病治療作用的研究

【摘要】:樺褐孔菌Inonotus obliquus (Fr.)Pilat是一種珍貴的藥用真菌,在我國主要分布在黑龍江、吉林省的大興安嶺地區(qū)。國外學者對樺褐孔菌的活性成分進行了研究,并將其用于腫瘤、糖尿病、心血管疾病、肝病及艾滋病的防治中。目前我國學者對樺褐孔菌的研究還處于起步階段。本課題首次對樺褐孔菌進行小鼠急性毒性實驗研究,通過研究樺褐孔菌提取物對小鼠實驗性Ⅱ型糖尿病機體氧化應(yīng)激狀態(tài)、慢性炎癥反應(yīng)以及糖脂代謝的影響,評價其藥效并進一步研究藥效物質(zhì)基礎(chǔ)。 采用STZ誘導(dǎo)實驗性Ⅱ型糖尿病小鼠模型,以二甲雙胍片、消渴靈片為陽性藥,研究樺褐孔菌水提物及醇提物對Ⅱ型糖尿病小鼠的影響。連續(xù)給藥4周,檢測動物飲水量、采食量、體重和FBG值。末次給藥后2h麻醉小鼠取血,用血細胞儀檢測血全細胞分類,用全自動生化分析儀檢鋇(?)TC、TG、HDL、LDL,用酶標儀檢測SOD、MDA、GSH-Px、 GSP、FINS,用ELISA法檢測(?)TNF-α、IL.8、IL-10等相關(guān)指標。結(jié)果表明:樺褐孔菌水提物及醇提物均能緩解實驗性Ⅱ型糖尿病小鼠的一般癥狀,降低FBG、FINS、GSP、TC、 TG、WBC和PLT,提高機體抗氧化能力,降低氧化產(chǎn)物MDA含量;抑制炎性反應(yīng),與模型對照組相比有差異顯著。 采用水提醇沉法制取樺褐孔菌多糖,結(jié)合光電甩尾實驗和小鼠體能評價實驗,考察樺褐孔菌多糖對糖尿病的綜合治療作用,結(jié)果表明樺褐孔菌多糖能降低Ⅱ型糖尿病小鼠FBG、GSP、FINS,減少自發(fā)活動次數(shù),縮短光電甩尾潛伏期,延長小鼠負重游泳力竭時間,與模型對照組相比有差異顯著。 結(jié)論:樺褐孔菌提取物對實驗性Ⅱ型糖尿病小鼠有顯著的治療作用,能提高小鼠體力和機體抗氧化能力,調(diào)節(jié)血脂代謝,抑制炎性反應(yīng),緩解外周神經(jīng)病變;水提物的作用優(yōu)于醇提物,其中水提物中的多糖成分有一定的降血糖作用,但其作用弱于水提物。
[Abstract]:Betula obliquus Inonotus obliquus (Fr.) Pilat is a precious medicinal fungus, which is mainly distributed in Heilongjiang and Daxing'anling area of Jilin Province. Foreign scholars have studied the active components of Betula obliquus and used them in the prevention and treatment of tumor, diabetes, cardiovascular disease, liver disease and AIDS. At present, the research on Betula obliquus is still in its infancy. In this study, the acute toxicity of Betula obliquus in mice was studied for the first time. The effects of the extract of Betula obliquus on oxidative stress, chronic inflammatory reaction and metabolism of glucose and lipid in mice with type 2 diabetes mellitus were studied. Evaluate its efficacy and further study its substance basis. The experimental model of type 2 diabetes induced by STZ was used to study the effects of water extract and alcohol extract of Betula platyphylla on type 2 diabetic mice with metformin tablets and Xiaokeling tablets as positive drugs. After 4 weeks of administration, the amount of water consumed, intake, body weight and FBG were measured. The blood samples were collected from anesthetized mice 2 hours after the last administration, the whole blood cell classification was detected by blood cell analyzer, the LDLs of TCG-TGG + HDLX, GSH-Px, GSPFINSwere detected by enzyme standard instrument, and the related indexes such as TNF- 偽 IL.8- IL-10 were detected by ELISA method. The results showed that the water extract and alcohol extract of Betula obliquus could relieve the general symptoms of experimental type 2 diabetic mice, decrease the Tc, TGG WBC and PLT, increase the antioxidant ability, decrease the content of MDA, and inhibit the inflammatory reaction. There was significant difference between the model control group and the model control group. The polysaccharides of Betula obliquus were prepared by water extraction and alcohol precipitation. The comprehensive therapeutic effect of polysaccharides from Betula obliquus on diabetes mellitus was investigated by combining the photoelectric tail flick test and the physical fitness evaluation test of mice. The results showed that the polysaccharides of Betula obliquus could decrease the number of spontaneous activities, shorten the latency of photoelectricity tail flick and prolong the exhausted time of swimming in mice with type 鈪,
