藥物及酒精對實驗性糖尿病大鼠腸系膜微循環(huán)的影響

【摘要】:目的:研究去甲腎上腺素(Norepinephrine、NE)、硝普鈉(Sodium nitroprusside、SNP)及酒精(Alcohol)對麻醉實驗性糖尿病大鼠腸系膜微動、靜脈血管反應(yīng)性的影響。 方法:正常雄性SD大鼠(220~250g)78只,誘導(dǎo)實驗性糖尿病大鼠(STZ)78只。采用BI-2000醫(yī)學(xué)圖像分析系統(tǒng),采集麻醉大鼠腸系膜微循環(huán)顯微圖像,比較觀察局部滴加不同濃度NE(10-12~10-4mol/L)、SNP(10-10~10-4mol)、酒精(0.025%~3.2%)及灌胃不同濃度酒精(0.375%~48%、5min)對實驗性糖尿病大鼠腸系膜微血管直徑的影響,并且觀察酚妥拉明(Phentolamine)對NE收縮血管作用的影響及左旋硝基精氨酸甲酯(L-NAME)對SNP舒張血管的影響。 1.第一部分,局部分別滴加不同濃度NE(10-12~10-4mol/L)于對照組及實驗性糖尿病大鼠腸系膜微血管,觀察NE對大鼠腸系膜微動、靜脈血管反應(yīng)性的影響;酚妥拉明孵育1min后,NE對大鼠腸系膜微動、靜脈血管收縮作用的影響。 2.第二部分,局部分別滴加不同濃度SNP(10-10~10-4mol/L)于對照組及實驗性糖尿病大鼠腸系膜微血管,觀察SNP對大鼠腸系膜微動、靜脈血管反應(yīng)性的影響;L-NAME孵育1min后,SNP對大鼠腸系膜微動、靜脈血管舒張作用的影響。 3.第三部分,局部滴加不同濃度酒精(0.025%~3.2%)及灌胃不同濃度酒精(0.375%~48%)對大鼠腸系膜微動、靜脈血管反應(yīng)性的影響。 結(jié)果: 1、第一部分,NE(10-12~10-4mol/L)濃度依賴性收縮大鼠腸系膜微動、靜脈直徑。NE誘導(dǎo)微動、靜脈收縮的濃度-效應(yīng)曲線的EC50(收縮達最大收縮50%時的藥物濃度)分別為(4.14±0.35)×10-9mol/L、(2.95±0.48)×10-8mol/L。與對照組相比較,實驗性糖尿病大鼠對NE的反應(yīng)性顯著減弱(P 0.05),NE誘導(dǎo)微動、靜脈收縮EC50分別為(5.39±0.29)×10-8mol/L、(4.78±0.39)×10-8mol/L。酚妥拉明孵育后,NE對對照組大鼠腸系膜微血管的收縮作用明顯強于STZ組。 2、第二部分,SNP(10-10~10-4mol/L)濃度依賴性舒張大鼠腸系膜微動、靜脈直徑,SNP誘導(dǎo)微動、靜脈舒張的EC50分別為(2.44±0.32)×10-8mol/L、(5.85±0.40)×10-8mol/L。與對照組相比較,,實驗性糖尿病大鼠腸系膜微血管對SNP的反應(yīng)性明顯減弱(P 0.05),SNP誘導(dǎo)微動、靜脈舒張EC50分別為(6.66±0.43)×10-8mol/L、(8.78±0.59)×10-8mol/L。L-NAME孵育后,SNP對對照組大鼠腸系膜微血管的舒張率明顯高于STZ組。 3、第三部分,酒精濃度依賴性舒張大鼠腸系膜微動、靜脈直徑(P 0.05),局部滴加酒精誘導(dǎo)微動、靜脈舒張EC50分別為(0.13±0.02)%、(0.17±0.02)%,灌胃酒精誘導(dǎo)微動、靜脈舒張的EC50分別為(2.81±0.22)%、(2.85±0.21)%。與對照組相比較,局部滴加酒精誘導(dǎo)微動、靜脈舒張的EC50分別為(0.25±0.03)%、(0.32±0.03)%,灌胃酒精誘導(dǎo)微動、靜脈舒張的濃度-效應(yīng)曲線的EC50分別為(2.79±0.23)%、(3.25±0.23)%,實驗性糖尿病大鼠腸系膜微血管對酒精的反應(yīng)性明顯減弱。 結(jié)論: 1. STZ誘導(dǎo)實驗性糖尿病大鼠腸系膜微循環(huán)對NE的反應(yīng)性顯著減弱。2.STZ誘導(dǎo)實驗性糖尿病大鼠腸系膜微循環(huán)對SNP的反應(yīng)性明顯減弱。3. STZ誘導(dǎo)實驗性糖尿病大鼠腸系膜微循環(huán)對酒精的反應(yīng)性明顯減弱。

2. in the second part, the mesenteric microvessels of the control group and experimental diabetic rats were separately added with different concentrations of SNP (10-10 ~ 10-4mol/L) in the control group and experimental diabetic rats. The effects of SNP on the mesenteric micromovement and venous reactivity of the rats were observed. After L-NAME incubation, the effect of SNP on the mesenteric micromovement and the vasodilatation of the rats was induced by SNP.
3. in the third part, the effects of different concentrations of alcohol (0.025% ~ 3.2%) and alcohol (0.375% ~ 48%) on the reactivity of the mesentery and venous reactivity of the rat mesentery (0.375% ~ 48%) were added.

STZ induced diabetic rats, and the reactivity of mesenteric microcirculation to alcohol was significantly weakened in experimental diabetic rats.
