【摘要】：目的研究孕早期七氟烷母體麻醉對子代大鼠認(rèn)知功能的影響,并對相關(guān)機(jī)制進(jìn)行初步探討。方法將27只7周齡SD雌性大鼠隨機(jī)分為實(shí)驗(yàn)組-1(n=9)、實(shí)驗(yàn)組-2(n=9)和對照組(n=9)。于孕5~7 d時(shí)進(jìn)行麻醉處理,實(shí)驗(yàn)組-1,實(shí)驗(yàn)組-2分別持續(xù)吸入1.8%七氟烷4,8 h,對照組僅吸入氧氣8 h,氧流量均為2 L·min-1。每組均隨機(jī)取6只母鼠所產(chǎn)子代SD大鼠,待出生后第30天開始進(jìn)行Morris水迷宮系統(tǒng)測定其認(rèn)知功能,用逆轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)(RT-PCR)及免疫組化法檢測海馬組織NR2B mRNA和HGN mRNA及蛋白表達(dá)情況。結(jié)果 Morris水迷宮系統(tǒng)測定結(jié)果顯示,3組逃避潛伏期、穿越平臺(tái)次數(shù)及在平臺(tái)象限停留時(shí)間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組-1NR2BmRNA和HGN mRNA分別為0.79±0.11,0.66±0.08,實(shí)驗(yàn)組-2NR2BmRNA和HGN mRNA分別為0.77±0.09,0.65±0.07,對照組NR2B mRNA和HGN mRNA分別為0.78±0.08,0.68±0.07。3組NR2B mRNA和HGN mRNA表達(dá)結(jié)果差異無統(tǒng)計(jì)學(xué)意義(P0.05)。實(shí)驗(yàn)組-1、實(shí)驗(yàn)組-2及對照組NR2B蛋白陽性細(xì)胞的平均灰度值分別為99.68±17.68,97.62±18.63,101.38±15.52,HGN蛋白陽性細(xì)胞的平均灰度值分別為86.49±22.04,89.14±20.15,92.15±22.51,3組子代SD大鼠海馬NR2B和HGN蛋白表達(dá)量差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論孕早期七氟烷母體麻醉對子代SD大鼠海馬NR2B mRNA和HGN mRNA表達(dá)及NR2B蛋白表達(dá)無影響,是否與七氟烷麻醉劑量相關(guān)尚需進(jìn)一步研究。
[Abstract]:Objective to study the effect of sevoflurane maternal anesthesia on the cognitive function of offspring rats in early pregnancy and to explore the related mechanism. Methods 27 7-week-old female SD rats were randomly divided into three groups: experimental group (n = 9), experimental group (n = 9) and control group (n = 9). Anesthetic treatment was performed on the 7th day of gestation. The experimental group (-1) and the experimental group (-2) inhaled 1.8% sevoflurane for 48 h, while the control group only inhaled oxygen for 8 h and the oxygen flow rate was 2 L min-1. In each group, 6 SD rats were randomly selected from their offspring, and their cognitive function was measured by Morris water maze system on the 30th day after birth. The expression of NR2B mRNA and HGN mRNA and protein in hippocampus were detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. Results the results of Morris water maze system showed that there was no significant difference in escape latency, crossing times and staying time in platform quadrant among the three groups (P0.05). The expression of NR2B mRNA and HGN mRNA were 0.79 鹵0.110.66 鹵0.08 in experimental group, 0.77 鹵0.09 鹵0.65 鹵0.07 in experimental group, 0.78 鹵0.080.68 鹵0.07.3 in control group respectively (P0.05). The average gray values of NR2B protein positive cells in experimental group -2 and control group were 99.68 鹵17.68, 97.62 鹵18.63101.38 鹵15.52 and 86.49 鹵22.04 鹵89.14 鹵20.15 鹵92.15 鹵22.51, respectively. There was no significant difference in the expression of NR2B and HGN protein in hippocampus of SD rats (P0.05). Conclusion the maternal anesthesia of sevoflurane has no effect on the expression of NR2B mRNA, HGN mRNA and NR2B protein in hippocampus of SD rats in early pregnancy.
