商陸皂苷甲對(duì)水負(fù)荷大鼠腎臟AQP2及AQP4表達(dá)的影響

【摘要】:目的:觀察商陸皂苷甲對(duì)水負(fù)荷大鼠腎臟水通道蛋白2(AQP2)及水通道蛋白4(AQP4)表達(dá)的調(diào)節(jié)作用,研究商陸皂苷甲利尿作用機(jī)制。方法:將50只雄性SD大鼠隨機(jī)分為空白對(duì)照組、氫氯噻嗪組及商陸皂苷甲高、中、低劑量組,腹腔注射給藥,氫氯噻嗪組給予氫氯噻嗪0.4 mg/kg,商陸皂苷甲高、中、低劑量組分別給5.2、2.6、1.3 mg/kg商陸皂苷甲,收集并測(cè)量給藥后6 h內(nèi)的尿量;水負(fù)荷實(shí)驗(yàn)結(jié)束后將大鼠麻醉處死,運(yùn)用免疫組化及Real Time-PCR技術(shù)檢測(cè)大鼠腎臟AQP2、AQP4蛋白及mRNA表達(dá)的變化。結(jié)果:與空白對(duì)照組比較,商陸皂苷甲高劑量組大鼠尿量顯著增多,腎臟AQP2、AQP4蛋白及mRNA的表達(dá)顯著下調(diào)(P0.05);商陸皂苷甲中、低劑量組尿量及腎臟AQP2、AQP4蛋白及mRNA表達(dá)差異無統(tǒng)計(jì)學(xué)意義。結(jié)論:商陸皂苷甲通過下調(diào)大鼠腎臟AQP2、AQP4蛋白及mRNA表達(dá),使水分重吸收降低,可能是其利尿作用的機(jī)制之一。
[Abstract]:Aim: to investigate the effects of pokeweed saponins A on the expression of aquaporin 2AQP2and aquaporin 4AQP4 in the kidney of water-loaded rats, and to study the mechanism of poke-saponin A diuretic action. Methods: fifty male Sprague-Dawley rats were randomly divided into control group, hydrochlorothiazide group and pokeweed saponins A high, middle and low dose groups, intraperitoneal injection of hydrochlorothiazide 0.4 mg / kg, pokeweed saponin A high, medium. The rats in the low dose group were given 5.2 ~ 2.6A 1.3 mg/kg pokeweed saponin A, and the urine volume was collected and measured within 6 hours after administration, and the rats were killed under anesthesia after the water loading experiment. The changes of AQP2AQP4 protein and mRNA expression in the rat kidney were detected by immunohistochemistry and Real Time-PCR technique. Results: compared with the blank control group, the urine volume of high dose group of pokeweed saponin A increased significantly, and the expression of AQP2AQP4 protein and mRNA in kidney decreased significantly (P 0.05), but there was no significant difference in urine volume and renal AQP2AQP4 protein and mRNA expression in low dose group. Conclusion: pokeweed saponin A can decrease water reabsorption by down-regulating the expression of AQP2AQP4 protein and mRNA in rat kidney, which may be one of the mechanisms of its diuretic effect.
