隔藥餅灸對功能性胃腸病大鼠CRF、NT的影響

【摘要】:目的:以功能性胃腸病(FIGDs)大鼠為研究對象,觀察隔藥餅灸對FIGDs大鼠的治療作用;并運用ELISA法檢測實驗大鼠促腎上腺激素釋放因子(CRF)與神經(jīng)降壓素(NT)的血清含量,以了解隔藥餅灸對功能性胃腸病大鼠CRF、NT的影響,進而探討隔藥餅灸是否通過調(diào)節(jié)FIGDs大鼠的CRF、NT水平而起到治療作用,以期為隔藥餅灸治療FIGDs的臨床應(yīng)用提供一定的實驗依據(jù)。 方法:60只SD大鼠隨機分為5組:空白組(A組)、模型組(B組)、隔藥餅灸組(C組)、杏仁核注射組(D組)、假處理組(E組);各組先進行適應(yīng)性飼喂。正式實驗開始后除A組正常飼喂,其余各組均接受FIGDs造模;采用慢性束縛應(yīng)激+過度疲勞+飲食失節(jié)的造模方法并在此基礎(chǔ)上加夾尾、搖晃以造成FIGDs模型。于每日開始造模前30min,C組捆綁固定后進行隔藥餅灸,取Ⅰ:肝俞、脾俞、足三里;Ⅱ:章門、期門、足三里兩組穴位交替施灸;B、D、E組則只捆綁不施灸,造模連續(xù)進行21日;第22日D組進行雙側(cè)杏仁核微量注射CNQX溶液,E組則接受雙側(cè)杏仁核微量注射生理鹽水;自第22日起所有實驗大鼠均自由吃食飲水4日;所有實驗大鼠于第26天麻醉后取材并將標(biāo)本送檢。標(biāo)本用ELISA法測CRH、NT的血清含量;光鏡下觀察胃腸組織的形態(tài)。分別于造模前、初次束縛后、正式實驗的第14、第21、第26日觀察各組大鼠一般情況;分別于造模前、造模結(jié)束、治療后測量各組大鼠體重、食量、尿D-木糖排泄率的變化并記錄。 結(jié)果:造模結(jié)束后,接受造模各組大鼠一般狀態(tài)變差,腹瀉便溏,體重、食量、尿D-木糖排泄率均低于空白組且差異均有統(tǒng)計學(xué)意義(P0.01)。治療后,接受造模各組各項指標(biāo)較造模結(jié)束時均有所回升,C、D組高于模型組且差異有統(tǒng)計學(xué)意義(P0.01),E組與模型組比較差異無統(tǒng)計學(xué)意義。經(jīng)光鏡觀察胃腸組織:空白組:結(jié)腸組織結(jié)構(gòu)完整,細(xì)胞狀態(tài)良好;隔藥餅灸組與CNQX組:結(jié)腸壁結(jié)構(gòu)完整,細(xì)胞排列與形態(tài)良好,未見炎癥表現(xiàn);而模型組與假處理組均有炎癥表現(xiàn)。C、D組CRF、NT水平高于B組,差異有統(tǒng)計學(xué)意義(P0.01)。 結(jié)論: 1.隔藥餅灸和CNQX注射均通過上調(diào)CRF、NT水平起到維護機體應(yīng)激調(diào)節(jié)功能,增加抗應(yīng)激能力及抗炎癥、保護細(xì)胞的作用,能改善實驗大鼠的應(yīng)激消耗狀態(tài),對功能性胃腸病起到治療作用。 2.隔藥餅灸和杏仁核微量注射CNQX對CRF、NT的調(diào)節(jié)作用可能均與杏仁核AMPA受體相關(guān)。
[Abstract]:Objective: to observe the therapeutic effect of herbal cake moxibustion on FIGDs rats in rats with functional gastrointestinal disease (FIGDs), and to detect the serum levels of adrenocorticotropic hormone releasing factor (CRFF) and neurotensin (NTN) in experimental rats by ELISA method. In order to understand the effect of herbal cake moxibustion on CRFN NT in rats with functional gastroenteropathy, and to explore whether the moxibustion of medicine-separated cake could play a therapeutic role by regulating the level of CRFN in FIGDs rats, the purpose of this study was to provide some experimental evidence for the clinical application of drug separated cake moxibustion in the treatment of FIGDs. Methods Sixty Sprague-Dawley rats were randomly divided into five groups: control group (group A), model group (group B), moxibustion group (group C), amygdala injection group (group D) and sham treatment group (group E). At the beginning of the formal experiment, all the other groups were fed with FIGDs except group A, and the FIGDs model was made by using the method of chronic restraint stress and excessive fatigue diet disjointing and adding tail clip to make FIGDs model.
