不同規(guī)格毫針針刺對(duì)大鼠穴區(qū)筋膜組織、細(xì)胞和膠原纖維的影響

【摘要】:目的:探討不同規(guī)格毫針針刺對(duì)大鼠腧穴穴區(qū)筋膜結(jié)締組織結(jié)構(gòu)、細(xì)胞活性、膠原纖維排列形態(tài)及含量的影響。方法:將32只SD大鼠隨機(jī)分為空白組、細(xì)針組、中針組和粗針組,每組8只。除空白組外,其他組分別采用不同規(guī)格毫針在大鼠"中脘"沿任脈循行方向平刺,施以捻轉(zhuǎn)平補(bǔ)平瀉刺激,每天1次,空白組予相同的麻醉、抓取、固定。干預(yù)3天后,切取各組穴區(qū)筋膜結(jié)締組織,采用HE染色、增殖細(xì)胞核抗原(proliferating cell nuclear antigen,PCNA)免疫組化染色和MASSON染色,鏡下觀察腧穴筋膜結(jié)締組織形態(tài)、細(xì)胞中PCNA表達(dá)、膠原纖維的排列形態(tài)和表達(dá)變化。結(jié)果:各組毫針針刺后,腧穴筋膜結(jié)締組織形態(tài)及膠原纖維排布一致性發(fā)生改變;各組腧穴筋膜結(jié)締組織細(xì)胞中PCNA蛋白表達(dá)均明顯增強(qiáng)(P0.01,P0.05);膠原纖維的面積分布發(fā)生改變,其中細(xì)針組較空白組增大,但差異無統(tǒng)計(jì)學(xué)意義(P0.05),中針組與粗針組膠原纖維的面積分布均較空白組減小(均P0.05)。結(jié)論:不同規(guī)格毫針針刺均能夠改變受治腧穴穴區(qū)局部組織形態(tài)、增強(qiáng)細(xì)胞活性和調(diào)節(jié)膠原纖維蛋白表達(dá)變化,這可能是毫針針刺治療"通經(jīng)脈"作用的細(xì)胞生物力學(xué)原理之一,但規(guī)格不同,其所產(chǎn)生的刺激量不同,穴區(qū)組織細(xì)胞和膠原纖維變化的量-效差異存在復(fù)雜關(guān)系。
[Abstract]:Objective: to investigate the effects of different kinds of millimeter acupuncture on connective tissue structure, cell activity, collagenous fiber arrangement and content of fascia in acupoints of rats. Methods: 32 Sprague-Dawley rats were randomly divided into three groups: blank group, fine needle group, middle needle group and coarse needle group, with 8 rats in each group. In addition to the blank group, the other groups were treated with the same anesthesia, grasping and fixing the same anesthesia as the blank group, respectively, by using different specifications of the needle in the direction of "Zhongwan" along the meridian of the rat, and applying twirling, tonifying, flattening and reducing stimulation once a day. After 3 days of intervention, the connective tissue of the fascia of the acupoints in each group was removed and stained with HE, proliferating cell nuclear antigen-PCNA (PCNA) and MASSON staining. The connective tissue morphology of acupoints and the expression of PCNA in the cells were observed under microscope. Changes in the morphology and expression of collagen fibers. Results: after acupuncture, the morphology of connective tissue and the arrangement consistency of collagenous fibers in acupoints were changed, the expression of PCNA protein in connective tissue cells of acupoints was significantly enhanced, and the area distribution of collagen fibers was changed. The area distribution of collagen fibers in the medium needle group and the coarse needle group were all decreased compared with the blank group (P 0.05), but there was no significant difference between the fine needle group and the blank group (P 0.05). Conclusion: acupuncture with different specifications can change the local tissue morphology, enhance cell activity and regulate the expression of collagen fibrin in acupoints. This may be one of the cellular biomechanical principles in the treatment of "Tongjing", but with different specifications and different stimuli, there is a complex relationship between the changes of tissue cells and collagen fibers in the acupoint area.
