【摘要】：目的探究鈣激活氯通道(Ca CCs)TMEM16A在大鼠心房和心室組織中的表達(dá)情況,為臨床治療心肌缺血、心律失常以及心力衰竭等疾病提供新的治療靶點(diǎn)。方法 2014年10月—2015年9月,將18只清潔級(jí)SpragueDawley大鼠適應(yīng)性飼養(yǎng)1周,采用10%水合氯醛溶液腹腔注射麻醉后取心臟,分離心房和心室組織進(jìn)行實(shí)驗(yàn)。采用Western blotting法和實(shí)時(shí)熒光定量反轉(zhuǎn)錄PCR(RT-q PCR)法分別檢測(cè)大鼠心房和心室組織中TMEM16A及其mRNA水平。結(jié)果大鼠心房和心室組織TMEM16A水平分別為(0.46±0.02)和(0.33±0.03),心房和心室組織TMEM16A mRNA水平分別為(3.55±0.17)和(1.00±0.03),大鼠心房組織中TMEM16A及其mRNA水平高于心室組織(P0.01)。結(jié)論 Ca CCs TMEM16A在大鼠心房和心室組織中均有表達(dá),其在心房組織中的水平高于心室組織。
[Abstract]:Objective to investigate the expression of Ca CCs)TMEM16A in rat atrium and ventricle, and to provide a new therapeutic target for the treatment of myocardial ischemia, arrhythmia and heart failure. Methods from October 2014 to September 2015, 18 clean grade SpragueDawley rats were fed adaptively for one week. The hearts were taken after intraperitoneal injection of 10% chloral hydrate solution to separate the atrial and ventricular tissues. The levels of TMEM16A and mRNA in rat atrium and ventricle were detected by Western blotting and real-time fluorescence quantitative reverse transcription (PCR(RT-q). Results the levels of TMEM16A in rat atrium and ventricle were 0.46 鹵0.02) and 0.33 鹵0.03m, respectively. The levels of TMEM16A mRNA in atrial and ventricular tissues were 3.55 鹵0.17) and 1.00 鹵0.03, respectively. The levels of TMEM16A and mRNA in rat atrial tissue were higher than those in ventricular tissue (P 0.01). Conclusion Ca CCs TMEM16A is expressed in rat atrium and ventricle, and the level of Ca CCs TMEM16A in atrial tissue is higher than that in ventricular tissue.
