【摘要】：目的觀察小半夏湯對順鉑化療大鼠腦干孤束核Fos蛋白表達的影響。方法雄性大鼠隨機分為正常對照組(10ml·kg~(-1)蒸餾水i.g.)、順鉑模型組(0.1ml·kg~(-1)蒸餾水i.g.)、昂丹司瓊治療組(昂丹司瓊2.6mg·kg~(-1),i.g.)、阿瑞吡坦治療組(阿瑞吡坦11.1mg·kg~(-1),i.g.)、小半夏湯治療組(小半夏湯1.6g·kg~(-1),i.g.),單籠飼養(yǎng)。模型組和各治療組6 mg·kg~(-1)順鉑i.p.造模。從造模前3 d開始,各組動物每天按照上述劑量分別灌胃相應藥物2次(間隔12 h)直至處死。注射順鉑后24 h、72 h深度麻醉大鼠,心臟灌注,取腦,冰凍切片,免疫組化技術觀察各組大鼠孤束核內Fos陽性細胞數(shù)。結果與空白對照組比較,注射順鉑后24 h、72孤束核的Fos陽性細胞數(shù)均顯著增加(P0.001),造模后24h時間點的增加程度大于72 h。小半夏湯治療組與順鉑模型組比較,兩個時間點Fos陽性細胞數(shù)均顯著降低(P0.001);與空白對照組比較,小半夏湯治療組Fos陽性細胞數(shù)有增加趨勢,但無統(tǒng)計學意義(P0.05);與兩種陽性藥組分別進行比較,小半夏湯治療組Fos陽性細胞數(shù)沒有顯著性差異(P0.05)。結論順鉑能夠使大鼠孤束核Fos蛋白表達顯著增加,小半夏湯可以抑制順鉑導致的孤束核Fos蛋白表達增加。
[Abstract]:Objective to observe the effect of Xiao Banxia decoction on the expression of Fos protein in brain stem solitary tract nucleus of cisplatin chemotherapy rats. Methods male rats were randomly divided into three groups: normal control group (n = 10 ml) and cisplatin model group (n = 0.1 ml 路kg ~ (-1) in distilled water (i.g.), Ondansetron treatment group (2.6mg / kg ~ (-1) i.g., Arelpidem treatment group (11.1mg kg ~ (-1), small Banxia decoction group (small Banxia decoction 1.6 g kg 路kg ~ (-1) i.g.), single cage feeding, and the control group (n = 10) were randomly divided into three groups: the control group (n = 10), the control group (n = 10); Model group and treatment group (6 mg 路kg ~ (-1) cisplatin i.p. Make a model. From the 3 days before the model was made, the animals in each group were given the corresponding drugs twice a day (12 hours interval) according to the above dosage until they were killed. The number of Fos positive cells in the nucleus tractus solitarii of rats in each group was observed by immunohistochemical technique. The rats were anesthetized for 72 hours after injection of cisplatin, the heart was perfused, brain was taken, frozen sections were taken and the number of Fos positive cells was observed by immunohistochemistry. Results compared with the control group, the number of Fos positive cells in the nucleus tractus solitarii at 24 h after injection of cisplatin increased significantly (P 0.001), and the increase at 24 h after injection was more than 72 h. Compared with cisplatin model group, the number of Fos positive cells in Xiaobanxia decoction group was significantly lower than that in cisplatin model group, and the number of Fos positive cells in Xiaobanxia decoction treatment group was increased compared with the blank control group. However, there was no significant difference in the number of Fos positive cells between the two groups, and there was no significant difference in the number of Fos positive cells between the two groups. Conclusion cisplatin can significantly increase the expression of Fos protein in rat nucleus of solitary tract, and Xiaobanxiatang can inhibit the expression of Fos protein in nucleus of solitary tract induced by cisplatin.
