芪芍顆粒治療橋本甲狀腺炎的實驗研究

【摘要】:目的:通過觀察芪芍顆粒對實驗性自身免疫性甲狀腺炎(EAT)模型大鼠血清甲狀腺球蛋白抗體(TGAb)、甲狀腺過氧化物酶抗體(TPOAb)、白介素-6(IL-6)、白介素-12(IL-12)的影響,探討芪芍顆粒防治橋本甲狀腺炎(HT)的機理。材料與方法:采用SPF級SD雌性大鼠60只,周齡4-6周,體質(zhì)量110±10g。具體方法如下:(1)第1周適應性喂養(yǎng);(2)第2周將大鼠隨機(采用隨機數(shù)表法)分為正常對照組(15只)、造模組(45只),正常對照組給予蒸餾水,造模組給予濃度為0.64g/L的高碘水;(3)第4周開始給予造模組大鼠雙足皮下多點注射由完全弗氏佐劑(CFA)充分乳化成油包水狀的Tg(Tg:CFA=1:1),一共2次,中間間隔2天,作為初次免疫;(4)第5周開始給予大鼠后背皮下多點注射由弗氏不完全免疫佐劑(IFA)乳化的Tg(Tg:IFA=1:1),一共4次,中間間隔7天,作為加強免疫;第8周結(jié)束時成模,將成模后的大鼠隨機(采用隨機數(shù)表法)分為模型組(15只)、雷公藤多苷片組(15只)、芪芍顆粒組(15只);(5)第9周各組開始灌胃給藥,雷公藤多苷片組每日給予雷公藤多苷片6.25mg/kg,芪芍顆粒組每日給予中藥21.4g/kg,模型組及正常對照組每日給予等量的生理鹽水;(6)灌胃治療4周后腹腔麻醉,經(jīng)腹主動脈取血離心并采用酶聯(lián)免疫法(ELISA法)來檢測各組大鼠血清中的IL-6、IL-12、TGAb、TPOAb的水平。結(jié)果:1.與正常對照組相比,模型組大鼠血清甲狀腺抗體TPOAb、TGAb的水平明顯升高,有顯著性差異(p0.01);與模型組相比,芪芍顆粒組大鼠血清甲狀腺抗體TPOAb、TGAb的水平明顯降低,有顯著性差異(p0.01);與雷公藤多苷片組相比,芪芍顆粒組大鼠血清甲狀腺抗體TPOAb、TGAb的水平無明顯變化,無顯著性差異(p0.05)。2.與正常對照組相比,模型組大鼠血清IL-6、IL-12的水平明顯升高,有顯著性差異(p0.01);與模型組相比,芪芍顆粒組大鼠血清IL-6、IL-12的水平明顯降低,有顯著性差異(p0.01);與雷公藤多苷片組相比,芪芍顆粒組大鼠血清IL-6、IL-12的水平無明顯變化,無顯著性差異(p0.05)。結(jié)論:1.芪芍顆?娠@著降低EAT大鼠血清甲狀腺抗體TGAb、TPOAb的水平。2.在降低EAT大鼠血清甲狀腺抗體TGAb、TPOAb的水平上,芪芍顆粒與雷公藤多苷片相比,無顯著性差異。3.芪芍顆粒能夠顯著降低EAT大鼠血清IL-6、IL-12的水平。4.在降低EAT大鼠血清IL-6、IL-12的水平上,芪芍顆粒與雷公多苷片相比,無顯著性差異。5.芪芍顆粒與雷公藤多苷片治療作用相當。
[Abstract]:Objective: To observe the effect of Qi Pai Granule on serum thyroglobulin antibody (TGAb), thyroid peroxidase antibody (TPOAb), interleukin -6 (IL-6) and -12 (IL-12) in experimental autoimmune thyroiditis (EAT) model rats, and to explore the mechanism of Qi Shao Granule on the prevention and control of Hashimoto's thyroiditis (HT). 60 rats of sex were 4-6 weeks of age, and the body mass of 110 + 10g. was as follows: (1) first weeks of adaptive feeding; (2) second weeks were randomly divided into normal control group (15) and model group (45), the normal control group was given distilled water and the model group was given the high iodine concentration of 0.64g/L; (3) the model rats began to be given to the model rats at week fourth. The multipoint injection of the bipedal subcutaneous injection was fully emulsified by the complete Freund's adjuvant (CFA) into a water like Tg (Tg:CFA=1:1), a total of 2 times, 2 days in the middle interval, as the primary immunization. (4) the rat's dorsal subcutaneous multipoint injection of Tg (Tg:IFA=1:1), which was emulsified by the Freund incomplete immune adjuvant (IFA), was given to the back of the rat's back, 4 times, for 7 days in the middle interval, as an enhanced immunization. At the end of the eighth week, the rat model was divided into model group (15 rats), the group of Tripterygium wilfordii (15) and Qi Shao granule group (15); (5) ninth weeks, each group began to give the medicine. The Tripterygium glycoside group was given the Tripterygium glycoside tablet 6.25mg/kg daily, and the Qi Pai granule group was given 21.4g/kg, the model group and the model group daily. The normal control group was given equal amount of normal saline daily; (6) intraperitoneal anesthesia after 4 weeks of gavage, blood centrifugation through abdominal aorta and enzyme linked immunoassay (ELISA) to detect the levels of IL-6, IL-12, TGAb and TPOAb in serum of each group. Results: 1. compared with the normal control group, the level of serum thyroid antibody TPOAb, TGAb level in the model group rats was compared with the normal control group. Significantly higher, there was significant difference (P0.01). Compared with the model group, the level of serum thyroid antibody TPOAb and TGAb in the rats of Qi Shao granule group decreased significantly (P0.01). Compared with the group of Tripterygium wilfordii polysaccharide, the level of serum thyroid antibody TPOAb, TGAb in the rats of Qi Shao granule group had no significant change, no significant difference (P0.05).2. and no significant difference. Compared with the normal control group, the level of serum IL-6 and IL-12 in the model group was significantly higher than that in the model group (P0.01). Compared with the model group, the serum IL-6, IL-12 level of the rats in the Qi Shao granule group decreased significantly (P0.01). Compared with the group of Tripterygium wilfordii polysaccharide, there was no significant change in the level of IL-6 and IL-12 in the serum of the rats of the Qi Pei granule group. There was no significant difference (P0.05). Conclusion: 1. Qi paehao granule can significantly reduce the serum thyroid antibody TGAb of EAT rats. The level.2. of TPOAb can reduce the serum thyroid antibody TGAb, TPOAb level of EAT rats. There is no significant difference between Qi Shao granule and Tripterygium wilfordii multi glycoside tablets, there is no significant difference between.3. Qi paehao granule and EAT rat serum IL-6, IL-12 water Flat.4. has no significant difference in the level of IL-6 and IL-12 in the serum of EAT rats. There is no significant difference between the Qi Pai granule and the Lei Gong poly glycoside tablets. The effect of.5. on the treatment is similar to that of Tripterygium wilfordii polysaccharide.
