異丙酚減輕氯胺酮麻醉對幼年大鼠海馬神經(jīng)元和認知功能的損害(英文)

【摘要】:目的探討異丙酚與氯胺酮復(fù)合麻醉對幼鼠發(fā)育中大腦認知功能的影響。方法 SD幼年大鼠被隨機分為生理鹽水組(NS組),氯胺酮組(K組);異丙酚組(P組),異丙酚+氯胺酮組(P+K組),每組20只,均腹腔注射相應(yīng)藥物1 ml,維持麻醉6 h。麻醉結(jié)束后各組幼鼠隨機分為相同數(shù)量的A、B兩部分,每部分10只。A部分于麻醉結(jié)束后取出幼鼠海馬組織制作石蠟切片,進行海馬CA1區(qū)神經(jīng)元凋亡和蛋白質(zhì)Bcl-2/Bax表達的分析;B部分于麻醉結(jié)束后3周行Morris水迷宮測試。結(jié)果 K組凋亡率較NS組明顯增加(P0.05);P+K組凋亡率較P組明顯上升(P0.05),但與K組相比,凋亡率明顯減少(P0.05)。P+K組Bcl-2/Bax蛋白相對表達明顯低于P組,但明顯高于K組(P0.01)。Morris水迷宮結(jié)果顯示K組學習記憶功能明顯差于NS組(P0.05);P組及P+K組學習記憶功能優(yōu)于K組(P0.05)。結(jié)論異丙酚復(fù)合氯胺酮麻醉,可明顯減輕氯胺酮所致的幼年大鼠海馬神經(jīng)元的凋亡和大鼠遠期認知功能的損害,異丙酚上調(diào)Bcl-2基因表達可能是其作用機制之一。
[Abstract]:Objective to investigate the effects of propofol and ketamine combined anesthesia on brain cognitive function in young rats.Methods Young SD rats were randomly divided into normal saline group (n = 20), ketamine group (n = 20), propofol group (n = 20), propofol group (n = 20) and propofol ketamine group (n = 20).After anesthesia, young rats in each group were randomly divided into two parts, the same number of Agna B, and 10 rats in each part were removed from the hippocampus after anesthesia to make paraffin sections.Apoptosis and protein Bcl-2/Bax expression in hippocampal CA1 neurons were analyzed. Morris water maze test was performed 3 weeks after anesthesia.Results the apoptotic rate of group K was significantly higher than that of group NS, but the relative expression of Bcl-2/Bax protein in group K was significantly lower than that in group P (P 0.05), but the relative expression of Bcl-2/Bax protein in group P (P 0.05) was significantly lower than that in group P (P < 0.05), but the expression of Bcl-2/Bax protein in group K was significantly lower than that in group P (P < 0.05).Conclusion propofol combined with ketamine anesthesia can significantly reduce the apoptosis of hippocampal neurons induced by ketamine and the long-term cognitive function in rats. The up-regulation of Bcl-2 gene expression by propofol may be one of its mechanisms.
