乳鐵蛋白對(duì)神經(jīng)病理性痛大鼠脊髓小膠質(zhì)細(xì)胞活化的影響研究

【摘要】:目的探討乳鐵蛋白對(duì)神經(jīng)病理性痛大鼠脊髓背角小膠質(zhì)細(xì)胞活化的影響。方法雄性SD大鼠32只,體質(zhì)量200~220 g,按照數(shù)字表法隨機(jī)分為4組(對(duì)照組、神經(jīng)病理性痛組、米諾環(huán)素組、乳鐵蛋白組),每組8只。對(duì)照組大鼠僅分離坐骨神經(jīng),不結(jié)扎,肌鞘內(nèi)注射生理鹽水8μl;其余3組大鼠采用結(jié)扎坐骨神經(jīng)的方法制備神經(jīng)病理性痛模型。神經(jīng)病理性痛組鞘內(nèi)僅注射生理鹽水8μl;乳鐵蛋白組鞘內(nèi)注射乳鐵蛋白100μg,米諾環(huán)素組鞘內(nèi)注射米諾環(huán)素(小膠質(zhì)細(xì)胞特異性活化抑制劑)100μg。給藥后每隔30 min以熱刺激法測(cè)試大鼠熱縮爪潛伏期,共180 min,測(cè)量7次后,常規(guī)處死大鼠取脊髓背角,采用免疫熒光法檢測(cè)小膠質(zhì)細(xì)胞特異性標(biāo)記物Iba-1的表達(dá),并進(jìn)行統(tǒng)計(jì)學(xué)分析。結(jié)果與對(duì)照組比較,神經(jīng)病理性痛組縮爪潛伏期明顯延長(zhǎng),差異有統(tǒng)計(jì)學(xué)意義(P0.05);與神經(jīng)病理性痛組相比,米諾環(huán)素組和乳鐵蛋白組縮爪潛伏期延長(zhǎng),差異有統(tǒng)計(jì)學(xué)意義(P0.05);而米諾環(huán)素組與乳鐵蛋白組爪潛伏期比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),可間接證明乳鐵蛋白可通過(guò)抑制脊髓小膠質(zhì)細(xì)胞活化減輕大鼠神經(jīng)病理性痛。與對(duì)照組比較,神經(jīng)病理性痛組、米諾環(huán)素組和乳鐵蛋白組脊髓背角Iba-1表達(dá)明顯減少,差異有統(tǒng)計(jì)學(xué)意義(P0.05);與神經(jīng)病理性痛組相比,米諾環(huán)素組和乳鐵蛋白組脊髓背角Iba-1表達(dá)也明顯減少,差異有統(tǒng)計(jì)學(xué)意義(P0.05);而米諾環(huán)素組與乳鐵蛋白組縮爪潛伏期比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05),可直接證明乳鐵蛋白可通過(guò)抑制脊髓小膠質(zhì)細(xì)胞活化減輕大鼠神經(jīng)病理性痛。結(jié)論乳鐵蛋白可通過(guò)抑制大鼠脊髓小膠質(zhì)細(xì)胞活化減輕神經(jīng)病理性痛,對(duì)臨床麻醉鎮(zhèn)痛過(guò)程有一定的指導(dǎo)價(jià)值。
[Abstract]:Objective to investigate the effect of lactoferrin on the activation of microglia in spinal dorsal horn of neuropathic pain rats.Methods 32 male Sprague-Dawley rats with a body weight of 200 ~ 220g were randomly divided into 4 groups (control group, neuropathic pain group, minocycline group, lactoferrin group, 8 rats in each group).In the control group, the sciatic nerve was isolated, not ligated, and 8 渭 l of normal saline was injected intrathecally into the muscle, while the other three groups were used to establish the neuropathic pain model by ligating the sciatic nerve.In neuropathic pain group, intrathecal injection of normal saline was only 8 渭 l, lactoferrin 100 渭 g in lactoferrin group and minocycline 100 渭 g in minocycline group.The latent period of thermal contraction of claw was measured by thermal stimulation every 30 min. After 7 times of measurement, the spinal dorsal horn of rats were routinely killed and the expression of Iba-1, a specific microglial marker, was detected by immunofluorescence.Statistical analysis was carried out.Results compared with the control group, the paw contraction latency in the neuropathic pain group was significantly prolonged (P 0.05), and the paw contraction latency in minocycline group and lactoferrin group was longer than that in the neuropathic pain group.The difference was statistically significant (P 0.05), but there was no significant difference between minocycline group and lactoferrin group in paw latency, which indirectly proved that lactoferrin could alleviate neuropathic pain by inhibiting the activation of spinal microglia in rats.Compared with the control group, the expression of Iba-1 in the spinal dorsal horn of the neuropathic pain group, minocycline group and lactoferrin group was significantly decreased (P 0.05) and compared with the neuropathic pain group.The expression of Iba-1 in spinal dorsal horn of minocycline group and lactoferrin group was also significantly decreased (P 0.05), while the paw contraction latency of minocycline group and lactoferrin group was higher than that of lactoferrin group.The results showed that lactoferrin could alleviate neuropathic pain by inhibiting the activation of spinal microglia.Conclusion lactoferrin can relieve neuropathic pain by inhibiting the activation of microglia in spinal cord of rats.
