間斷重復(fù)七氟醚麻醉對(duì)新生大鼠學(xué)習(xí)記憶遠(yuǎn)期影響的研究

【摘要】:目的:本實(shí)驗(yàn)從行為學(xué)角度探討哺乳期大鼠間斷重復(fù)吸入臨床麻醉濃度和亞麻醉濃度的七氟醚對(duì)其幼年期、成年期學(xué)習(xí)記憶能力的影響。 方法:健康7d齡雄性SD大鼠24只,按照隨機(jī)區(qū)組化實(shí)驗(yàn)設(shè)計(jì),隨機(jī)分為三組(n=8),實(shí)驗(yàn)對(duì)照組(C組)、兩種不同濃度1.5%、2.6%七氟醚組(S1、S2組)。S1、S2組分別于出生第7、14、21d分三次各吸入七氟醚1h,C組于相同的時(shí)間單純吸入運(yùn)載氣體(1L/min O2+1L/min Air)1h。記錄每次吸入時(shí)S1、S2組大鼠麻醉生效時(shí)間、麻醉恢復(fù)時(shí)間及每次吸入麻醉前、麻醉生效時(shí)、麻醉生效30min時(shí)、麻醉恢復(fù)時(shí)的呼吸次數(shù)。各組大鼠出生第28d(P28)開始幼年期Morris水迷宮實(shí)驗(yàn),為期9d,由可視平臺(tái)實(shí)驗(yàn)、定位航行實(shí)驗(yàn)和空間探索實(shí)驗(yàn)3部分組成。P28、P29行可視平臺(tái)實(shí)驗(yàn),記錄游泳速度和運(yùn)動(dòng)總路程;可視平臺(tái)實(shí)驗(yàn)結(jié)束后,休息2d,于P32~P36行定位航行實(shí)驗(yàn),記錄逃避潛伏期、運(yùn)動(dòng)總路程,搜索策略;P36行空間探索實(shí)驗(yàn),記錄原平臺(tái)象限滯留時(shí)間百分比、運(yùn)動(dòng)距離百分比、進(jìn)入次數(shù)百分比及平臺(tái)區(qū)域滯留時(shí)間、運(yùn)動(dòng)距離、進(jìn)入次數(shù)。各組大鼠自然喂養(yǎng)至成年,于P91開始進(jìn)行成年期Morris水迷宮實(shí)驗(yàn),,為期14d,由定位航行實(shí)驗(yàn)、空間探索實(shí)驗(yàn)和不固定平臺(tái)實(shí)驗(yàn)3部分組成。于P91~P95行定位航行實(shí)驗(yàn),P95行空間探索實(shí)驗(yàn),實(shí)驗(yàn)方法及記錄指標(biāo)同幼年期;于P96~P104行不固定平臺(tái)實(shí)驗(yàn),為期9d,記錄達(dá)到標(biāo)準(zhǔn)所進(jìn)行的實(shí)驗(yàn)次數(shù)及所用的總時(shí)間。 結(jié)果:⑴麻醉生效時(shí)間與麻醉恢復(fù)時(shí)間:S1組3次麻醉生效時(shí)間(min)分別為6、7、9,麻醉恢復(fù)時(shí)間(min)分別為12、7、6;S2組3次麻醉生效時(shí)間(min)分別為4、6、8,麻醉恢復(fù)時(shí)間(min)分別為31、15、12。 ⑵吸入過程中呼吸次數(shù):三次吸入S1、S2組大鼠麻醉生效時(shí)及麻醉生效30min時(shí)呼吸次數(shù)比麻醉前明顯減少(P0.01);麻醉前與麻醉恢復(fù)后各組大鼠呼吸次數(shù)的差異無統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑶可視平臺(tái)實(shí)驗(yàn)游泳速度及總距離:與P28相比,各組大鼠總距離P29顯著增大(P0.01);各組大鼠游泳速度差異無統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑷定位航行實(shí)驗(yàn)搜索策略構(gòu)成比:不同測試日各組大鼠搜索策略構(gòu)成比差異有顯著統(tǒng)計(jì)學(xué)意義(P0.01),隨著測試天數(shù)增加,隨機(jī)式、邊緣式搜索逐漸減少;直線式、趨向式搜索逐漸增多。 ⑸定位航行實(shí)驗(yàn)逃避潛伏期:幼年期各組大鼠組間比較第1、2、4、5d S2組逃避潛伏期較C組增大(P0.05),第1、4、5d S1組逃避潛伏期較C組增大(P0.05),第2d S2組逃避潛伏期較S1組增大(P0.05);組內(nèi)比較S1、S2組第3d起逃避潛伏期縮短,C組第4d起逃避潛伏期縮短(P0.05)。成年期各組大鼠組間比較第4d S1組逃避潛伏期較C組延長(P0.05);組內(nèi)比較S2組、C組第5d逃避潛伏期縮短(P0.05),S1組各天間差異無統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑹定位航行實(shí)驗(yàn)運(yùn)動(dòng)總距離:幼年期各組大鼠組間比較第2、4、5d S2組運(yùn)動(dòng)總距離較C組增大(P0.05),第2d S2組運(yùn)動(dòng)總距離較S1組增大(P0.05);組內(nèi)比較S2組第3d起運(yùn)動(dòng)總距離縮短(P0.05),S1組、C組第2d起運(yùn)動(dòng)總距離縮短(P0.05)。成年期各組大鼠組間、組內(nèi)比較差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑺空間探索實(shí)驗(yàn)原平臺(tái)象限:幼年期各組大鼠原平臺(tái)象限滯留時(shí)間百分比、運(yùn)動(dòng)距離百分比及進(jìn)入次數(shù)百分比組間比較無統(tǒng)計(jì)學(xué)差異(P0.05)。成年期S1組原平臺(tái)象限滯留時(shí)間百分比22.07±6.93(%)較C組31.52±4.53(%)減小,差異有統(tǒng)計(jì)學(xué)意義(P0.05);S1組原平臺(tái)象限運(yùn)動(dòng)距離百分比20.20±4.95(%)較C組28.96±6.57(%)減小,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑻空間探索實(shí)驗(yàn)原平臺(tái)區(qū)域:幼年期各組大鼠原平臺(tái)區(qū)域滯留時(shí)間、運(yùn)動(dòng)距離及進(jìn)入次數(shù)組間比較差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。成年期S1組原平臺(tái)區(qū)域進(jìn)入次數(shù)1.86±0.90(次)較C組3.63±1.69(次)減少,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。 ⑼不固定平臺(tái)實(shí)驗(yàn):各組大鼠達(dá)到標(biāo)準(zhǔn)的實(shí)驗(yàn)次數(shù)(次)及達(dá)到標(biāo)準(zhǔn)所用的總時(shí)間(s)組間比較均無統(tǒng)計(jì)學(xué)差異(P>0.05)。 結(jié)論: ⑴哺乳期間斷重復(fù)吸入七氟醚可能損害大鼠幼年期的陳述性記憶能力,并存在濃度依賴性;而對(duì)空間聯(lián)想和回憶探究能力無影響; ⑵哺乳期間斷重復(fù)吸入臨床麻醉濃度七氟醚可能對(duì)成年期聯(lián)合型學(xué)習(xí)記憶能力及工作記憶能力無影響; ⑶哺乳期間斷重復(fù)吸入亞麻醉濃度七氟醚可能會(huì)損害大鼠成年期聯(lián)合型學(xué)習(xí)記憶能力,而對(duì)成年期工作記憶能力無影響。
[Abstract]:Objective: To explore the effect of sevoflurane on the learning and memory ability of young rats during adulthood.
