白豆蔻揮發(fā)油對(duì)糖尿病腎病大鼠腎臟保護(hù)作用

【摘要】:目的:研究白豆蔻揮發(fā)油對(duì)糖尿病腎病大鼠腎臟保護(hù)作用及其機(jī)制。方法:46只Wistar大鼠隨機(jī)分為正常組(6只)和模型組(40只),模型組以腹腔注射STZ建立糖尿病腎病模型,造模后隨機(jī)分模型組,白豆蔻低、高劑量組,六味地黃丸組4組,各10只。連續(xù)灌胃給藥21d,每周檢測(cè)1次血糖;最后一次給藥24h后麻醉大鼠腹主動(dòng)脈取血,檢測(cè)血清肌酐(Cre)、尿素氮(BUN)、白蛋白(ALB)含量;光鏡下觀察腎臟組織病理改變;免疫組化法檢測(cè)基質(zhì)金屬蛋白酶2(MMP-2)、轉(zhuǎn)化生長(zhǎng)因子β1(TGF-β1)、胰島素樣生長(zhǎng)因子2(IGF-2)的表達(dá)。結(jié)果:與正常組比較,模型組血糖顯著升高(P0.01),經(jīng)過(guò)給藥后各給藥組血糖不同程度下降,其中白豆蔻高劑量組血糖顯著下降(P0.05,P0.01);與模型組比較,各給藥組Cre、BUN、ALB含量均降低,其中白豆蔻高劑量組作用明顯(P0.05,P0.01)。光鏡下病理形態(tài)學(xué)觀察顯示,各給藥組對(duì)大鼠腎組織損傷有不同程度的改善作用;各給藥組MMP-2、TGF-β1、IGF-2蛋白表達(dá)均上調(diào)。結(jié)論:白豆蔻揮發(fā)油對(duì)腎臟有一定的保護(hù)作用,作用機(jī)制可能與上調(diào)MMP-2、TGF-β1、IGF-2的表達(dá)有關(guān),從而改善鏈脲佐菌素所致糖尿病腎病的病理改變。
[Abstract]:Objective: to study the protective effect and mechanism of essential oil of cardamom cardamom on the kidney of diabetic nephropathy rats. Methods: 46 Wistar rats were randomly divided into normal group (n = 6) and model group (n = 40). Diabetic nephropathy model was established by intraperitoneal injection of STZ in the model group. After the model was established, the rats were randomly divided into model group, low dose group, high dose group and Liuwei Dihuang pill group, 10 rats in each group. Blood glucose was measured once a week for 21 days, and blood was taken from abdominal aorta of anesthetized rats 24 hours after the last administration. Serum creatinine (creatinine), urea nitrogen bun (bun), albumin (ALBB) were detected; pathological changes of renal tissue were observed under light microscope; expression of matrix metalloproteinase 2 (MMP-2), transforming growth factor 尾 1 (TGF- 尾 1), insulin-like growth factor 2 (IGF-2) was detected by immunohistochemical method. Results: compared with normal group, The blood glucose of the model group increased significantly, and the blood glucose of each drug group decreased in varying degrees after administration, among which, the blood glucose of the high dose group decreased significantly (P0.05P0.01A), and compared with the model group, the ALB content of creatine BUNA in each administration group decreased, and the blood glucose of each group decreased in different degrees, compared with the model group, the blood glucose in the high dose group decreased significantly. Among them, the high dose group of cardamom had obvious effect on P0.05 and P0.01.The histopathologic observation under light microscope showed that each group had different degree of ameliorative effect on the renal tissue injury of rats. Conclusion: the essential oil of cardamom has a protective effect on the kidney, and the mechanism may be related to the up-regulation of the expression of MMP-2TGF- 尾 1IGF-2, thus improving the pathological changes of streptozotocin induced diabetic nephropathy.
