經(jīng)方小柴胡湯對(duì)小鼠肝纖維化的改善作用及其機(jī)制的研究

【摘要】:目的:探究經(jīng)方小柴胡湯對(duì)肝纖維化改善作用的機(jī)制。方法:將雄性C57BL/6小鼠隨機(jī)分為3組,即正常對(duì)照組、模型組和實(shí)驗(yàn)組。模型組和實(shí)驗(yàn)組小鼠腹腔注射二甲基亞硝胺(DMN),每周3次,實(shí)驗(yàn)組注射DMN的同時(shí)每天灌胃小柴胡湯。4周后麻醉處死各組小鼠,取小鼠血漿和肝臟,對(duì)肝臟進(jìn)行HE和masson染色;利用ELISA檢測(cè)試劑盒測(cè)定血清中白細(xì)胞介素1β(IL-1β)、白細(xì)胞介素6(IL-6)、腫瘤壞死因子α(TNF-α)和C反應(yīng)蛋白(CRP)以及肝臟4-羥脯氨酸(4-Hyp)含量;采用realtime PCR和western blot檢測(cè)肝臟IL-1β、IL-6、TNF-α、I型膠原蛋白(Collagen I)和α-平滑肌動(dòng)蛋白α(α-SMA)表達(dá)量、核因子κB抑制蛋白(IκB)的磷酸化水平及核因子κB(NF-κB)核積累量。結(jié)果:結(jié)果表明,實(shí)驗(yàn)組肝纖維化病理變化相比于模型組得到顯著改善(P0.05);實(shí)驗(yàn)組血清IL-1β、IL-6、TNF-α、CRP及肝臟4-Hyp與模型組相比其含量均明顯下降,且差異顯著(P0.05);實(shí)驗(yàn)組肝臟IL-1β、IL-6、TNF-α、Collagen Ⅰ和α-SMA的表達(dá)水平、IκB的磷酸化水平及NF-κB的核積累量相比于模型組顯著下降(P0.05)。結(jié)論:小柴胡湯能顯著改善小鼠肝纖維化,其作用機(jī)制與抑制肝臟內(nèi)的炎癥反應(yīng)有關(guān)。
[Abstract]:Objective: to explore the mechanism of improving liver fibrosis by Meridian Xiaochaihu decoction. Methods: male C57BL / 6 mice were randomly divided into three groups: normal control group. Model group and experimental group. Mice in model group and experimental group were intraperitoneally injected with Dimethylnitrosamine (DMN) 3 times a week. Mice in the experimental group were anesthetized and killed by intragastric administration of Xiaochaihu decoction for 4 weeks, and plasma and liver were taken from mice. The liver was stained with HE and masson, and the contents of IL-1 尾, IL-6, TNF- 偽 and C-reactive protein (CRP- 偽) and 4 hydroxyproline (Hyp4) in the liver were determined by ELISA assay kit. Realtime PCR and western blot were used to detect the expression of IL-1 尾 -IL-6TNF- 偽 Collagen I, 偽 -SMAA, phosphorylation of nuclear factor 魏 B inhibitor I 魏 B, and nuclear accumulation of nuclear factor 魏 B (NF- 魏 B) NF- 魏 B. results: the results showed that the expression of 偽 -smooth muscle actin 偽 (偽 -SMAA), the phosphorylation level of nuclear factor- 魏 B inhibitor (NF- 魏 B) and the nuclear accumulation of NF- 魏 B in the liver were measured by realtime PCR and western blot. Compared with the model group, the pathological changes of liver fibrosis in the experimental group were significantly improved (P 0.05), the serum levels of IL-1 尾 -IL-6 TNF- 偽 -hr and the contents of liver 4-Hyp in the experimental group were significantly lower than those in the model group. The level of phosphorylation of I 魏 B and the nuclear accumulation of NF- 魏 B in the experimental group were significantly lower than those in the model group. Conclusion: Xiaochaihu decoction can significantly improve liver fibrosis in mice, and the expression level of TNF- 偽 Collagen 鈪,
