玫參顆粒對圍絕經(jīng)期大鼠內(nèi)分泌-免疫功能的影響

【摘要】:目的:探討玫參顆粒治療婦女更年期綜合征的內(nèi)在機(jī)制。方法:大鼠隨機(jī)分為7組,即假手術(shù)組、模型組、坤寶丸陽性對照組、玫參顆粒10g/kg組、玫參顆粒5g/kg組、玫參顆粒2.5g/kg組、戊酸雌二醇片組。除了假手術(shù)外,其他各組去除雙側(cè)卵巢造模,各給藥組給予相應(yīng)藥物灌胃,模型組與假手術(shù)組給予蒸餾水灌胃,連續(xù)干預(yù)30d,腹腔麻醉后腹主動(dòng)脈取血,測定大鼠血清雌二醇(E2)、卵泡刺激素(FSH)、促黃體生成激素(LH)、超氧化物歧化酶(SOD)和白介素2(IL-2)的水平,處死大鼠后計(jì)算大鼠子宮、胸腺、脾臟、腎上腺系數(shù);HE染色觀察子宮、陰道組織形態(tài)學(xué)變化。結(jié)果:(1)玫參顆粒5g/kg組、10g/kg組能明顯抑制因大鼠去卵巢而造成的體重增加,其中以10g/kg組的抑制幅度有極顯著差異。(2)玫參顆粒能顯著提高去卵巢大鼠子宮、腎上腺系數(shù),其中10g/kg組提高幅度有極顯著差異。(3)玫參顆粒能顯著提高去卵巢大鼠胸腺和脾臟系數(shù),其中2.5g/kg組、5g/kg組、10g/kg組提高胸腺系數(shù)幅度均有極顯著差異;5g/kg組提高脾臟系數(shù)有顯著性差異,10g/kg組提高脾臟系數(shù)有極顯著差異。(4)玫參顆粒能提高去卵巢大鼠血清中SOD、IL-2的水平,其中2.5g/kg組、5g/kg組、10g/kg組提高SOD水平幅度有極顯著地差異;2.5g/kg組、5g/kg組提高IL-2水平有顯著性意義,10g/kg組提高IL-2幅度有極顯著差異。(5)玫參顆粒能提高雙側(cè)卵巢切除的wistar雌性大鼠血清E2水平,2.5g/kg組提高幅度達(dá)顯著水平,5g/kg組、10g/kg組提高幅度達(dá)極顯著水平;玫參顆粒還能顯著降低去卵巢大鼠升高的FSH水平和LH水平,2.5g/kg組、5g/kg組、10g/kg組均能極顯著降低FSH水平,2.5g/kg組能顯著降低去卵巢大鼠LH水平,5g/kg組、10g/kg組能極顯著降低去卵巢大鼠LH水平。(6)通過子宮、陰道HE染色發(fā)現(xiàn)玫參顆粒能顯著改善去卵巢大鼠子宮內(nèi)徑狹小、萎縮的現(xiàn)象,2.5g/kg組、5g/kg組、10g/kg組改善均有顯著性差異;玫參顆粒還能改善陰道萎縮癥狀,2.5g/kg組、5g/kg組、10g/kg組改善均具有顯著性差異。結(jié)論:玫參顆粒治療圍絕經(jīng)期綜合征與調(diào)節(jié)機(jī)體內(nèi)分泌-免疫系統(tǒng)有關(guān),為臨床應(yīng)用和新藥開發(fā)提供實(shí)驗(yàn)數(shù)。
[Abstract]:Objective: to explore the intrinsic mechanism of Meishen granule in the treatment of menopausal syndrome in women. Methods: rats were randomly divided into 7 groups: sham operation group, model group, Kun Bao pill positive control group, Meishen granule 10 g / kg group, Meishen granule 5 g / kg group, Meishen granule 2.5 g / kg group. Estradiol valerate group: except sham-operation, the other groups removed the bilateral ovarian model, each group was given the corresponding drugs by stomach, the model group and the sham operation group were given distilled water for 30 days, the abdominal aorta blood was taken after abdominal anaesthesia, and the rats in the model group were treated with distilled water for 30 days. The serum levels of estradiol E _ (2), follicle stimulating hormone (FSH), luteinizing hormone (LHH), superoxide dismutase (SOD) and interleukin-2 (IL-2) were measured. The uterine, thymus, spleen and adrenal gland coefficients were observed by HE staining after the rats were killed. Results the weight gain caused by ovariectomized rats was significantly inhibited in 5g / kg Meisen granule group, and the inhibition range of 10g / kg group was very significant difference (P < 0.05). Meishen granule could significantly increase the uterus of ovariectomized rats. The coefficient of adrenal gland in 10 g / kg group was significantly higher than that in 10 g / kg group (P < 0.05). Meisen granule could significantly increase the coefficients of thymus and spleen in ovariectomized rats. There were significant differences in the amplitude of thymus coefficient in 2.5 g / kg group and 5 g / kg group in 10 g / kg group. There were significant differences in the spleen coefficient of 5 g / kg group. The increase of spleen coefficient in 10 g / kg group could increase the level of IL-2 in serum of ovariectomized rats. There was significant difference in the increase of SOD level in 2.5 g / kg group and 5 g / kg group in 10 g / kg group. There was significant difference between 2.5 g / kg group and 5 g / kg group in increasing IL-2 level. There was a significant difference in increasing IL-2 amplitude in 10 g / kg group.) Meishen granule could increase the level of IL-2 in bilateral ovariectomized wistar female rats. The serum E _ 2 level was significantly increased in 2.5 g / kg group, and the increase range was extremely significant in 10 g / kg group of 5 g / kg group. Meishen granule also significantly decreased the elevated FSH level in ovariectomized rats and LH level in 2.5 g / kg group and 10 g / kg group in 5 g / kg group. The FSH level was significantly decreased in 2.5 g / kg group. The LH level in ovariectomized rats was significantly decreased in 5 g / kg group and 10 g / kg group in 10 g / kg group. Rat LH level. 6) through the uterus, Vagina HE staining showed that Meishen granules could significantly improve the narrow uterine diameter and atrophy in the uterus of ovariectomized rats. There were significant differences in the improvement of uterine diameter and atrophy in the 2.5 g / kg group and the 5 g / kg group (10 g / kg group). Meisen granules can also improve the symptoms of vaginal atrophy. There are significant differences in the improvement of the symptoms of vaginal atrophy in the 5 g / kg group and 10 g / kg group. Conclusion: the treatment of peri-menopausal syndrome by Meisen granule is related to regulating the endocrine and immune system of the body. To provide experimental data for clinical application and new drug development.
