【摘要】：目的：本課題通過(guò)建立慢性心衰大鼠模型,觀察養(yǎng)心湯對(duì)慢性心衰大鼠血清內(nèi)皮素-1(ET-1)的水平和左室短軸縮短率(LVFS)的影響,以及心肌組織病理學(xué)的改變,探討?zhàn)B心湯治療慢性心衰大鼠的作用機(jī)制。方法：將60只SD大鼠隨機(jī)選取10只作為空白對(duì)照組,其余50只用阿霉素連續(xù)腹腔注射進(jìn)行造模,6周后將造模成功的大鼠隨機(jī)平均分為3組,每組14只,進(jìn)行灌胃給藥。養(yǎng)心湯組給予養(yǎng)心湯懸濁液劑量2.34g/kg/d灌胃；卡托普利組給予卡托普利懸濁液劑量2.25mg/kg/d灌胃；空白對(duì)照組與模型對(duì)照組給予蒸餾水2ml/d灌胃。4周后,給予大鼠腹腔注射水合氯醛(3.5ml/kg)麻醉,使用超聲診斷儀進(jìn)行心功能測(cè)定后,取血5m1用以檢測(cè)血清內(nèi)皮素1的濃度,取大鼠心肌組織染色進(jìn)行病理觀察。結(jié)果：1.與模型對(duì)照組比較,養(yǎng)心湯組和卡托普利組大鼠的血清ET-1濃度水平有所下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05)；養(yǎng)心湯組與卡托普利組相比,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。2.與模型對(duì)照組比較,養(yǎng)心湯組和卡托普利組大鼠的LVFS有所增加,差異有統(tǒng)計(jì)學(xué)意義(P0.05)；養(yǎng)心湯組與卡托普利組比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。3.與模型對(duì)照組比較,養(yǎng)心湯組和卡托普利組均能減輕心衰大鼠心肌組織的病理?yè)p傷。結(jié)論：1.養(yǎng)心湯能夠降低慢性心衰大鼠血清ET-1的水平,其作用與卡托普利相當(dāng)。2.養(yǎng)心湯能夠提高心衰大鼠的LVFS,其作用與卡托普利相當(dāng)。3.養(yǎng)心湯對(duì)慢性心衰大鼠心肌損傷組織有一定的修復(fù)作用,并且與卡托普利相當(dāng)。
[Abstract]:Objective: to observe the effect of Yangxin decoction on serum endothelin-1 (ET-1) and left ventricular shortening rate (LVFSs) in rats with chronic heart failure (CHF), and to observe the pathological changes of myocardium in rats with chronic heart failure (CHF). To explore the mechanism of Yangxin decoction in the treatment of chronic heart failure rats. Methods: 60 SD rats were randomly selected as the blank control group. The other 50 rats were randomly divided into 3 groups with 14 rats in each group after 6 weeks of continuous intraperitoneal injection of adriamycin. The captopril group was given a dose of 2.25 mg / kg / d of captopril suspension, the blank control group and the model control group were given 2 ml / d distilled water for 4 weeks, then the rats were anesthetized by intraperitoneal injection of chloral hydrate (3.5 ml / kg), and the cardiac function was measured by ultrasonic diagnostic apparatus. The concentration of serum endothelin 1 was detected by blood 5m1, and the myocardial tissue staining of rats was taken for pathological observation. Results compared with the model control group, the serum ET-1 level in the Yangxin decoction group and captopril group was lower than that in the model control group. There was no significant difference between Yangxin decoction group and captopril group. Compared with model control group, the LVFS of Yangxin decoction group and captopril group was increased. There was no significant difference between Yangxin decoction group and captopril group, and there was no significant difference between Yangxin decoction group and captopril group. There was no significant difference between Yangxin decoction group and captopril group, and there was no significant difference between Yangxin decoction group and captopril group. Both Yangxin decoction group and captopril group can reduce the pathological damage of myocardial tissue in rats with heart failure. Conclusion: 1. Yangxin decoction can reduce the level of serum ET-1 in chronic heart failure rats. The effect of Yangxin decoction was similar to that of captopril. 2. Yangxin decoction could improve LVFSof heart failure rats, and its effect was similar to that of captopril. Yangxin decoction had certain repair effect on myocardial injury tissue of chronic heart failure rats, and was similar to captopril.
