不同時(shí)間的乳化氟碳保存液處理對(duì)供體肺抗炎抗氧化能力及組織結(jié)構(gòu)的影響

【摘要】:目的:觀察乳化氟碳保存液(FCE)和UW液處理供體肺在不同時(shí)間對(duì)肺組織抗炎、抗氧化能力及其結(jié)構(gòu)的影響.方法:選擇48只約350~400 g SD雄性大鼠隨機(jī)分為兩組(FCE組與UW組),FCE組與UW組分別又隨機(jī)各分為兩個(gè)亞組.對(duì)每只大鼠進(jìn)行麻醉、氣管插管與機(jī)械通氣后,剖胸,全身肝素化后將灌注管自大鼠右心室插入肺動(dòng)脈,再用保存液進(jìn)行灌注,灌注完后快速取下心肺組織置入相應(yīng)的保存液中進(jìn)行保存.乳化氟碳組(FCE)用乳化氟碳進(jìn)行灌注與保存,一組保存6 h(FCE-6 h),另一組保存12 h(FCE-12 h).UW液組(UW)用UW液灌注與保存,保存方法及保存時(shí)間同F(xiàn)CE組.在相應(yīng)時(shí)段取肺組織,檢測(cè)白細(xì)胞介素-1β(IL-1β)、白細(xì)胞介素-6(IL-6)、腫瘤壞死因子-α(TNF-α)、丙二醛(MDA)、髓過(guò)氧化酶(MPO)、超氧化物歧化酶(SOD)的水平并觀察其組織結(jié)構(gòu)變化.結(jié)果:(1)兩組保存液處理6 h,肺組織MDA、MPO含量及病理積分顯著低于12 h組(P0.05),SOD活性則顯著高于12 h組(P0.05);(2)FCE組兩個(gè)時(shí)段肺組織MPO含量及病理積分顯著低于UW組(P0.05);(3)FCE組在病理改變較輕微,而UW組則較重.結(jié)論:與UW液相比,乳化氟碳同樣具有抑制炎癥,延緩并減輕缺血保存期肺損傷的能力,且乳化氟碳在兩個(gè)時(shí)段的抗氧化能力均強(qiáng)于UW液.
[Abstract]:Objective: to observe the anti-inflammatory effects of emulsified fluorocarbon preservative fluid (FCEE) and UW solution on lung tissue at different time. Methods: 48 male SD rats were randomly divided into two groups: FCE group and UW group. After tracheal intubation and mechanical ventilation, thoracotomy and heparinization of the whole body inserted the perfusion tube into the pulmonary artery from the right ventricle of the rat. After perfusion, the heart and lung tissues were quickly removed and stored in the corresponding preservation solution. The emulsified fluorocarbon group was perfused and preserved with emulsified fluorocarbon, one group was stored for 6 h, the other group was stored for 12 h, the other group was perfused and preserved with UW solution. The method and time of preservation are the same as those of FCE group. The levels of interleukin-1 尾, interleukin-6 (IL-6), tumor necrosis factor- 偽 (TNF- 偽), malondialdehyde (MDAA), myeloperoxidase (MPO) and superoxide dismutase (SOD) were detected and the changes of tissue structure were observed. The content of MPO and the pathological score of lung tissue in 12 h group were significantly lower than those in UW group (P 0.05) and the pathological score was significantly lower than that in UW group (P 0.05) and the pathological score was significantly lower than that in UW group (P 0.05) and the pathological score was significantly lower than that in UW group (P 0.05) and the pathological score was significantly lower than that in UW group. Conclusion: compared with UW solution, emulsified fluorocarbon also has the ability of inhibiting inflammation, delaying and alleviating lung injury during ischemic preservation, and the antioxidant ability of emulsified fluorocarbon in both periods is stronger than that in UW solution.
