【摘要】：背景:獲取脂肪術(shù)中需要應(yīng)用不同的局部腫脹麻醉液,其可能會(huì)對(duì)細(xì)胞生物特性產(chǎn)生一定的影響。目的:探討體外培養(yǎng)條件下不同局部腫脹麻醉液對(duì)人脂肪干細(xì)胞增殖與成脂分化的影響。方法:取第3代人脂肪干細(xì)胞分別用布比卡因、羅哌卡因、利多卡因局部腫脹麻醉液進(jìn)行培養(yǎng),對(duì)照組用低糖DMEM培養(yǎng)基進(jìn)行培養(yǎng),檢測各組干預(yù)不同時(shí)間人脂肪干細(xì)胞增殖活性、細(xì)胞上清乳酸脫氫酶活性和成脂分化比例。結(jié)果與結(jié)論:不同局部腫脹麻醉液干預(yù)后1,2,3,4,5 d細(xì)胞上清乳酸脫氫酶活性值顯著高于對(duì)照組(P0.05),布比卡因組顯著高于羅哌卡因組、利多卡因組(P0.05)。各局部腫脹麻醉液組細(xì)胞生長吸光度值均低于對(duì)照組(P0.05),且布比卡因組顯著低于羅哌卡因組、利多卡因組(P0.05)。各組人脂肪干細(xì)胞成脂分化比例差異無顯著性意義(P0.05)。結(jié)果表明,局部腫脹麻醉液(布比卡因、羅哌卡因、利多卡因)均能夠抑制人脂肪干細(xì)胞增殖,但不會(huì)對(duì)細(xì)胞成脂分化產(chǎn)生明顯的影響。
[Abstract]:Background: different local swelling anesthetics are needed to obtain fat. Objective: to investigate the effects of different local swelling anesthetic fluids on the proliferation and adipogenic differentiation of human adipose stem cells in vitro. The third generation of human adipose stem cells were treated with bupivacaine. Ropivacaine and lidocaine were cultured in local swelling anaesthesia, while those in control group were cultured in low glucose DMEM medium. The proliferation activity of human adipose stem cells was detected in each group. Results and conclusion: after intervention of different local swelling anesthetic solution, lactic dehydrogenase activity and adipogenic differentiation ratio of the supernatant were 1 ~ 2o _ 3 ~ (4). The activity of lactate dehydrogenase in the supernatant of 5 days was significantly higher than that in the control group (P 0.05), and that in the bupivacaine group was significantly higher than that in ropivacaine group. The cell growth absorbance of lidocaine group was lower than that of control group, and that of bupivacaine group was significantly lower than ropivacaine group. There was no significant difference in adipogenic differentiation of human adipose stem cells in lidocaine group (P 0.05). The results showed that bupivacaine and ropivacaine were locally swollen anesthetic fluid. Lidocaine can inhibit the proliferation of human adipose stem cells, but has no obvious effect on adipogenic differentiation.
