肺氣腫小鼠Treg細(xì)胞中組蛋白去乙;富钚詫(duì)Treg/Th17細(xì)胞的作用及機(jī)制初步探討

目的:煙草煙霧暴露下建立小鼠肺氣腫模型,觀察調(diào)節(jié)性T細(xì)胞(regulatory T cell,Treg)中組蛋白去乙;福╤istone deacetylase,HDAC)活性對(duì)Treg/Th17細(xì)胞的平衡變化,探討吸煙導(dǎo)致肺氣腫中存在CD4+T細(xì)胞亞群之間相互失衡的機(jī)制。方法:將健康雄性8周齡Balb/c小鼠按照隨機(jī)數(shù)字法分為對(duì)照組、煙熏組,每組各30只,對(duì)照組暴露于新鮮空氣中自然喂養(yǎng)24周,煙熏組用香煙煙霧暴露24周建立小鼠肺氣腫模型。(1)蘇木精-伊紅(HE)染色觀察小鼠肺組織形態(tài)學(xué)的改變,計(jì)算兩組肺泡平均內(nèi)襯間隔(Mean linear intercepts,MLI)。(2)流式細(xì)胞術(shù)檢測(cè)小鼠外周血、脾臟和肺組織中CD4+、CD4+IL-17、CD4+IFN-γ、CD4+Foxp3+細(xì)胞的表達(dá)。(3)采用免疫磁珠法分選肺組織中Treg細(xì)胞,實(shí)時(shí)熒光定量PCR法(Real-time polymerase chain reaction,RT-PCR)...
