目的:應(yīng)用生物信息學(xué)方法對哮喘患者上氣道基因表達(dá)譜芯片進(jìn)行分析,進(jìn)一步探討其可能的發(fā)病機(jī)制。方法:從GEO（Gene expressionomnibus）數(shù)據(jù)庫下載哮喘表達(dá)譜的芯片數(shù)據(jù)（GSE41861）,利用R語言Limma軟件包篩選哮喘患者與非哮喘患者鼻黏膜的差異表達(dá)基因（differentially expressed genes,DEGs）,并對差異基因作基因本體論（gene ontology,GO）和京都基因與基因組百科全書（kyoto encyclopedia of genes and genomes,KEGG）通路富集分析。利用STRING在線數(shù)據(jù)庫進(jìn)行蛋白-蛋白相互作用（protein protein interaction,PPI）分析,并用Cytoscape軟件及其插件NetworkAnalyzer進(jìn)行可視化,尋找關(guān)鍵（Hub）基因。結(jié)果:共篩選出118個(gè)差異基因,其中包括93個(gè)上調(diào)基因和25個(gè)下調(diào)基因。對差異基因進(jìn)行生物信息學(xué)分析,上調(diào)DEGs主要參與唾液分泌相關(guān)通路和結(jié)核相關(guān)通路,下調(diào)DEGs主要參與TGF-β相關(guān)信號通路。蛋白網(wǎng)絡(luò)分析篩選出CDC5L、AR和A...
