哮喘患者上氣道基因表達譜芯片的生物信息學分析

  目的:應(yīng)用生物信息學方法對哮喘患者上氣道基因表達譜芯片進行分析,進一步探討其可能的發(fā)病機制。方法:從GEO(Gene expressionomnibus)數(shù)據(jù)庫下載哮喘表達譜的芯片數(shù)據(jù)(GSE41861),利用R語言Limma軟件包篩選哮喘患者與非哮喘患者鼻黏膜的差異表達基因(differentially expressed genes,DEGs),并對差異基因作基因本體論(gene ontology,GO)和京都基因與基因組百科全書(kyoto encyclopedia of genes and genomes,KEGG)通路富集分析。利用STRING在線數(shù)據(jù)庫進行蛋白-蛋白相互作用(protein protein interaction,PPI)分析,并用Cytoscape軟件及其插件NetworkAnalyzer進行可視化,尋找關(guān)鍵(Hub)基因。結(jié)果:共篩選出118個差異基因,其中包括93個上調(diào)基因和25個下調(diào)基因。對差異基因進行生物信息學分析,上調(diào)DEGs主要參與唾液分泌相關(guān)通路和結(jié)核相關(guān)通路,下調(diào)DEGs主要參與TGF-β相關(guān)信號通路。蛋白網(wǎng)絡(luò)分析篩選出CDC5L、AR和A...
