神經(jīng)酰胺對胎鼠肺泡Ⅱ型上皮細(xì)胞增殖的影響

【摘要】:取1只妊娠17.5 d的ICR小鼠,將其體內(nèi)的胎鼠肺泡Ⅱ型上皮細(xì)胞(AECⅡ)經(jīng)分離、純化、培養(yǎng)、鑒定后,分別加入不同濃度的神經(jīng)酰胺,培養(yǎng)12、24和48 h后進(jìn)行MTT試驗(yàn)。在Transwell上構(gòu)建AECⅡ單層,以10μmol/L的神經(jīng)酰胺與AECⅡ培養(yǎng)12和24 h,并設(shè)置相應(yīng)對照組,應(yīng)用伏歐計(jì)測定跨上皮電阻值。結(jié)果顯示,隨著神經(jīng)酰胺染毒濃度的升高和染毒時間的延長,AECⅡ細(xì)胞增殖抑制率呈上升趨勢。10μmol/L的神經(jīng)酰胺分別刺激融合的AECⅡ單層12和24 h后,與未處理對照組比較,神經(jīng)酰胺刺激組的跨上皮電阻值顯著下降。
