核轉(zhuǎn)錄因子Nrf2與肺部炎癥疾病研究進(jìn)展

【摘要】:核因子NF-E2相關(guān)因子(nuclear factor erythroid 2-related factor 2,Nrf2)是氧化應(yīng)激反應(yīng)敏感基因的重要轉(zhuǎn)錄因子之一,可調(diào)節(jié)依賴抗氧化反應(yīng)元件(antioxidant response elements,ARE)的多種抗氧化蛋白的生成。研究顯示,Nrf2-Keap1(Kelch樣ECH聯(lián)合蛋白1)-ARE信號(hào)通路是細(xì)胞內(nèi)重要的內(nèi)源性抗氧化防御機(jī)制之一,上調(diào)該信號(hào)通路可誘導(dǎo)其下游調(diào)控的多種抗氧化酶及解毒酶,提高谷胱甘肽及超氧化物歧化酶等抗氧化物質(zhì)的表達(dá)水平,清除自由基,維持細(xì)胞內(nèi)氧化還原平衡狀態(tài),發(fā)揮內(nèi)源性細(xì)胞保護(hù)作用。近年來(lái),大氣污染尤其PM2.5可能是導(dǎo)致呼吸道炎癥發(fā)病率升高以及炎癥加重的關(guān)鍵因素之一,Nrf2及其信號(hào)通路對(duì)氣道炎癥性疾病如支氣管哮喘、慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)和急性肺損傷(acute lung injury,ALI)的調(diào)控,有望成為防治肺部炎癥性疾病藥物研究潛在性靶點(diǎn)。本文就Nrf2與氣道炎癥和氧化應(yīng)激相關(guān)信號(hào)通路調(diào)控進(jìn)行綜述。
[Abstract]:Nuclear factor NF-E2 related factor (nuclear factor erythroid 2-related factor 2n rf2 is one of the important transcription factors of oxidative stress response sensitive genes. It can regulate the production of many antioxidant proteins which depend on the antioxidant response element (antioxidant response elements,ARE). It has been shown that Nrf2-Keap1 (Kelch like ECH Associated protein 1)-ARE signaling pathway is one of the important endogenous antioxidant defense mechanisms in cells, and up-regulation of the signaling pathway can induce a variety of antioxidant enzymes and detoxifying enzymes regulated downstream. To improve the expression level of glutathione and superoxide dismutase (SOD), scavenge free radicals, maintain redox equilibrium in cells, and play an endogenous role in cell protection. In recent years, air pollution, especially PM2.5, may be one of the key factors leading to the increase of respiratory inflammation and the exacerbation of inflammation. Nrf2 and its signaling pathway play an important role in airway inflammatory diseases such as bronchial asthma. The regulation of chronic obstructive pulmonary disease (chronic obstructive pulmonary disease,COPD) and acute lung injury (acute lung injury,ALI) is expected to be a potential target for drug research in the prevention and treatment of pulmonary inflammatory diseases. This review reviews the regulation of Nrf2 and airway inflammation and oxidative stress related signaling pathways.
