正常組與人工模擬寒環(huán)境組大鼠肺組織蛋白組學(xué)差異分析

【摘要】:目的:探討正常組與人工模擬寒環(huán)境組大鼠肺組織蛋白組學(xué)差異表達(dá)及質(zhì)譜分析。方法:選取20只SD大鼠隨機(jī)分為正常對(duì)照組與寒環(huán)境組,每組10只。寒環(huán)境組模型復(fù)制使用人工氣候箱:(T)=(6±2)℃,每天刺激4 h(其余時(shí)間為正常組溫濕度);正常組(T)=(21±2)℃,每天持續(xù)24 h。造模7 d后,兩組大鼠于麻醉前12 h禁食、禁水,稱量體重。麻醉后開胸取出左肺組織10 mg/只,每組取3個(gè)樣本進(jìn)行樣品蛋白質(zhì)制備、雙向電泳實(shí)驗(yàn)、圖像分析、質(zhì)譜檢測(cè)。結(jié)果:對(duì)兩組差異蛋白圖譜進(jìn)行蛋白質(zhì)組學(xué)分析,鑒定出9個(gè)蛋白在兩組之間差異表達(dá),寒環(huán)境組與正常對(duì)照組比較,其中4個(gè)表達(dá)上調(diào)、5個(gè)表達(dá)下調(diào)。結(jié)論:通過(guò)蛋白質(zhì)組學(xué)分析,鑒定出的蛋白與外寒環(huán)境有相關(guān)性。
