骨髓間充質(zhì)干細(xì)胞移植聯(lián)合吡菲尼酮治療肺間質(zhì)纖維化的研究

【摘要】:目的探討吡菲尼酮(PFD)聯(lián)合骨髓間充質(zhì)干細(xì)胞(MSCs)移植入博來(lái)霉素(BLM)誘導(dǎo)肺纖維化小鼠的療效。方法 36只健康清潔級(jí)體重200 g雄性SD大鼠,隨機(jī)分為6組,A組為陰性對(duì)照組、B組陽(yáng)性對(duì)照組、C組單純激素治療組、D組單純MSCs移植組、E組單純PFD治療組、F組PFD聯(lián)合MSCs移植組,各6只。SD大鼠體外培養(yǎng)MSCs,注入BLM制備肺纖維化模型,觀察肺組織形態(tài)學(xué)變化,測(cè)定轉(zhuǎn)化生長(zhǎng)因子(TGF-β1)的蛋白表達(dá)。結(jié)果造模7d后,C、D、E、F組大鼠出現(xiàn)炎癥細(xì)胞浸潤(rùn)比B組減少,肺纖維化程度明顯減輕,F組較C、D、E組的病理改變更輕,肺纖維化不明顯。肺系數(shù)中B組下降明顯,與A組比較差異有統(tǒng)計(jì)學(xué)意義(P0.05)。B、C、D、E、F組肺纖維化評(píng)分高于A組,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。肺組織中轉(zhuǎn)化生長(zhǎng)因子膠原表達(dá)的陽(yáng)性細(xì)胞數(shù)明顯減少,F組較B、C、D、E組數(shù)量減少和強(qiáng)度減弱更加明顯,A組與B、C、D、E、F組比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05),C、D、E組間差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論骨髓間充質(zhì)干細(xì)胞移植聯(lián)合毗菲尼酮對(duì)肺纖維化有一定的抑制作用,能夠降低肺組織轉(zhuǎn)化生長(zhǎng)因子的表達(dá),有修復(fù)和再生作用,治療肺纖維化效果明顯。
[Abstract]:Objective to investigate the effect of pifeinone (PFD) combined with bone marrow mesenchymal stem cell (MSCs) transplantation into bleomycin (BLM) induced pulmonary fibrosis in mice. Methods Thirty-six healthy, clean weight 200g male SD rats were randomly divided into 6 groups: group A was negative control group, group B was positive control group, group C was treated with hormone alone, group D was treated with MSCs, group E was treated with PFD alone, group F was treated with MSCs. Six SD rats were cultured with MSCs, in vitro to make pulmonary fibrosis model. The changes of lung histomorphology and the expression of TGF- 尾 1 protein were observed. Results after 7 days, the inflammatory cell infiltration was decreased and the degree of pulmonary fibrosis was alleviated. The pathological changes in group F were lighter than those in group C, and the pulmonary fibrosis was not obvious. The pulmonary coefficient of group B was significantly lower than that of group A (P0.05). The score of pulmonary fibrosis in group B was higher than that in group A (P0.05). The number of positive cells of transforming growth factor collagen expression in lung tissue was significantly decreased, and the number and intensity of TGF- collagen positive cells in group F was significantly lower than that in group B (P 0.05). The difference between group A and group F was statistically significant (P0.05), but there was no significant difference between group C and C (P0.05). Conclusion Bone marrow mesenchymal stem cell transplantation combined with pifinidone can inhibit pulmonary fibrosis, reduce the expression of transforming growth factor in lung tissue, repair and regenerate, and have obvious effect in the treatment of pulmonary fibrosis.
