【摘要】：目的探討貝母素甲(PM)對(duì)脂多糖(LPS)誘導(dǎo)小鼠急性肺損傷過(guò)程中炎癥因子表達(dá)的影響。方法 80只昆明種小鼠體重(20±5)g隨機(jī)分成正常組、模型組、地塞米松(Dex)組、PM組,腹腔注射戊巴比妥(40 mg/kg)麻醉小鼠,模型組、Dex組及PM組小鼠給予氣管內(nèi)注射4 mg/kg LPS誘導(dǎo)建立急性肺損傷模型。注入LPS前1、12、24 h和36 h和注入后12 h,24 h和36 h,模型組、Dex組、PM組動(dòng)物分別給予生理鹽水、10 mg/kg鹽酸地塞米松、1 mg/kg PM溶液,正常組給予同等體積生理鹽水。實(shí)驗(yàn)結(jié)束后解剖肺臟,觀察肺臟形態(tài)學(xué)改變,分別用Real-Time PCR和酶聯(lián)免疫吸附試驗(yàn)(ELISA)測(cè)定腫瘤細(xì)胞壞死因子(TNF)-α、白介素(IL)-2、IL-6和IL-8。結(jié)果 LPS能明顯引起小鼠肺部的炎癥反應(yīng),PM處理后拮抗了LPS對(duì)小鼠肺組織的破壞作用,抑制促炎因子TNF-α、IL-2、IL-6、IL-8的表達(dá)。結(jié)論 PM對(duì)急性肺損傷小鼠肺組織保具有保護(hù)作用,其機(jī)制可能與下調(diào)炎癥因子的表達(dá)有關(guān)。
[Abstract]:Objective to investigate the effect of (PM) on the expression of inflammatory factors during acute lung injury induced by lipopolysaccharide (LPS) in mice. Methods 80 Kunming mice with body weight (20 鹵5) g were randomly divided into normal group, model group, dexamethasone (Dex) group and PM group. The mice were anesthetized by intraperitoneal injection of pentobarbital (40 mg/kg). Models of acute lung injury were induced by intratracheal injection of 4 mg/kg LPS in model group and PM group. The rats in the model group were given 10 mg/kg dexamethasone hydrochloride 1 mg/kg PM solution for 10 mg/kg in the model group and the same volume of normal saline in the normal group after injection of LPS for 24 h and 36 h and 12 h and 36 h after the injection, and the rats in the model group were given 10 mg/kg dexamethasone hydrochloride (Dexamethasone) 1 mg/kg PM solution, respectively. At the end of the experiment, the lung was dissected and the changes of lung morphology were observed. Tumor necrosis factor (TNF)-偽, interleukin (IL)-2-6 and IL-8 were measured by Real-Time PCR and enzyme linked immunosorbent assay (ELISA) respectively. Results LPS could obviously induce the inflammatory response in lung of mice treated with PM. It antagonized the destruction of lung tissue by LPS and inhibited the expression of IL-6 and IL-8 of TNF- 偽 and IL-2- 偽. Conclusion PM has protective effect on lung tissue protection in mice with acute lung injury, and its mechanism may be related to down-regulation of inflammatory factor expression.
