紅景天對哮喘小鼠氣道炎癥和氣道重塑的影響

【摘要】:目的:探討紅景天對哮喘小鼠氣道炎癥和氣道重塑的調(diào)控作用及機(jī)制。方法:40只C57bL/6小鼠隨機(jī)分成對照組、哮喘組、布地奈德組、紅景天低、高劑量組(0.21,1 g/kg),每組8只。小鼠分別于第0、14天以O(shè)VA致敏,第24天開始每天霧化吸入2.5%OVA激發(fā)并持續(xù)18 d,建立哮喘模型。紅景天組致敏后每次激發(fā)前0.5 h予以紅景天灌胃。劉氏染色法分類計(jì)數(shù)支氣管肺泡灌洗液(BALF)中細(xì)胞數(shù),肺組織病理學(xué)檢查觀察呼吸道炎癥、膠原染色面積,ELISA法檢測BALF中TGF-β1、IL-4和IFN-γ的濃度,免疫組化法檢測肺組織中TGF-β1、IL-4和IFN-γ的表達(dá)。結(jié)果:與對照組相比較,哮喘組小鼠呼吸道炎癥、膠原染色面積明顯加重,肺組織中IL-4、IL-4/IFN-γ、TGF-β1升高,IFN-γ降低(P0.05);布地奈德和紅景天能減輕小鼠氣道炎癥及膠原染色面積,降低肺組織中IL-4、IL-4/IFN-γ、TGF-β1,升高IFN-γ(P0.05)。結(jié)論:紅景天能夠抑制哮喘小鼠氣道炎癥和氣道重塑,其機(jī)制可能是通過調(diào)節(jié)IL-4/IFN-γ失衡,抑制TGF-β1的表達(dá)有關(guān)。
[Abstract]:Objective: to investigate the effect and mechanism of Rhodiola on airway inflammation and airway remodeling in asthmatic mice. Methods 40 C57bL/6 mice were randomly divided into control group, asthma group, budesonide group, low salidroid group and high dose group (0.21 g/kg) with 8 mice in each group. The mice were sensitized with OVA on day 0 and day 14, respectively, and the asthma model was established by daily inhalation of 2.5%OVA for 18 days from day 24. Rhodiola group was sensitized 0.5 h before each stimulation. The number of cells in bronchoalveolar lavage fluid (BALF) was counted by Liu's staining method, the respiratory tract inflammation was observed by pathological examination of lung tissue, the concentration of TGF- 尾 _ (1) IL-4 and IFN- 緯 in BALF was detected by Elisa and the expression of TGF- 尾 _ (1) IL-4 and IFN- 緯 in lung tissue was detected by immunohistochemistry. Results: compared with the control group, the airway inflammation and collagen staining area in asthmatic mice were significantly increased, IL-4 / IFN- 尾 1 increased and IFN- 尾 1 decreased (P0.05), budesonide and Rhodiola could reduce airway inflammation and collagen staining area in mice. Decrease IL-4 / IFN- 緯 TGF- 尾 1 and increase IFN- 緯 in lung tissue (P0.05). Conclusion: Rhodiola can inhibit airway inflammation and airway remodeling in asthmatic mice by regulating IL-4 / IFN- 緯 imbalance and inhibiting the expression of TGF- 尾 _ 1.
