霧化吸入布地奈德干預(yù)肺纖維化大鼠及其機(jī)制的實(shí)驗(yàn)研究

【摘要】:目的:探討霧化吸入布地奈德對博萊霉素誘導(dǎo)的大鼠肺纖維化的影響及其可能機(jī)制。方法:45只SD雌性大鼠隨機(jī)分為3組(各組15只):對照組、模型組和干預(yù)組,肺纖維化模型行氣管內(nèi)滴入博萊霉素造模。干預(yù)組造模后第7天霧化吸入布地奈德,模型組和對照組霧化吸入等量生理鹽水,霧化后第7天、第14天、第28天各組隨機(jī)處死5只動(dòng)物并分別保存肺組織,HE和Masson染色判斷肺組織肺泡炎及肺纖維化程度;免疫組化染色測定肺組織轉(zhuǎn)化生長因子-β(transforming growth factor-β,TGF-β)、結(jié)締組織生長因子(connective tissue growth factor,CTGF)的表達(dá)水平;RT-PCR檢測Ⅰ、Ⅲ型膠原mRNA的表達(dá)水平。結(jié)果:肺組織病理學(xué)觀察顯示對照組肺泡炎和肺纖維化程度明顯輕于模型組和干預(yù)組,干預(yù)組較模型組有明顯改善;模型組和干預(yù)組肺組織中TGF-β和CTGF水平較對照組均有明顯增高(P0.05),但干預(yù)組較模型組表達(dá)減少(P0.05);模型組Ⅰ、Ⅲ型膠原表達(dá)明顯高于對照組(P0.05),而干預(yù)組較模型組表達(dá)明顯減少(P0.05)。結(jié)論:布地奈德具有明顯的抗纖維化作用,機(jī)制可能為通過抑制細(xì)胞因子的表達(dá)而減少肺組織中Ⅰ、Ⅲ型膠原的生成,最終抑制肺纖維化。
[Abstract]:Aim: to investigate the effect of inhaled budesonide on bleomycin induced pulmonary fibrosis in rats and its possible mechanism. Methods 45 female Sprague-Dawley rats were randomly divided into three groups (15 rats in each group): control group, model group and intervention group. Pulmonary fibrosis model was made by intratracheal instillation of bleomycin. In the intervention group, budesonide was inhaled on the 7th day after modeling, and the same amount of saline was inhaled by the model group and the control group, and on the 7th day and the 14th day after the atomization, budesonide was inhaled into the model group. On the 28th day, 5 animals were killed randomly and the lung tissues were preserved by HE and Masson staining to determine the degree of pulmonary alveolitis and pulmonary fibrosis. The expression of transforming growth factor- 尾 (TGF- 尾) and connective tissue growth factor (connective tissue growth factor- 尾 (CTGF- 尾) in lung tissue were detected by immunohistochemistry. Results: the pulmonary histopathological observation showed that the degree of alveolitis and pulmonary fibrosis in the control group was significantly less than that in the model group and the intervention group, and the intervention group was significantly improved compared with the model group.
