雙氫青蒿素對百草枯致小鼠肺纖維化的治療研究

【摘要】:目的:探討雙氫青蒿素(dihydroartemisin,DHA)對急性百草枯(paraquat,PQ)染毒小鼠的治療作用。方法:將134只小鼠隨機分成3組:正常對照組(n=24)、PQ染毒組(n=55,實際存活24只)和DHA治療組(n=55,實際存活35只)。PQ組與DHA組:以100mg/kg PQ一次性灌胃;2h后DHA組以20mg/kg DHA灌胃治療,PQ組與對照組灌胃等量蒸餾水,每日1次。觀察給藥后第3,7,14,21天小鼠的一般情況,比較生存率差異,計算肺系數(shù),酶聯(lián)免疫吸附法(ELISA)測定肺組織TGF-β1的表達;HE及Masson染色觀察小鼠肺組織病理變化。結(jié)果:PQ組21d生存率與對照組和DHA組比較差異有統(tǒng)計學意義(P0.05)。PQ組肺組織中TGF-β1在第3,7,14,21天分別為(467.77±46.52)ng/L、(434.89±48.94)ng/L、(1 410.70±94.07)ng/L、(2 306.89±33.06)ng/L;DHA組一般情況較PQ組好,TGF-β1表達在第3,7,14,21天時分別為(441.54±23.58)ng/L、(443.22±46.83)ng/L、(750.49±29.96)ng/L、(1 554.95±92.30)ng/L,較PQ組差異顯著(P0.01)。隨時間推移,PQ組肺泡炎加重,14d始伴隨著TGF-β1的升高,肺組織出現(xiàn)纖維化改變,以第21天時最為明顯,與對照組比較差異明顯(P0.01)。DHA組炎性細胞浸潤較PQ組少,充血水腫輕,肺纖維化程度較PQ組減輕(P0.05)。結(jié)論:DHA可能通過降低肺組織中TGF-β1表達,發(fā)揮對PQ中毒小鼠肺纖維化的治療作用。
[Abstract]:Objective: to investigate the therapeutic effect of dihydroartemisinine dihydroartemisinin (DHA) on mice exposed to acute paraquat PQ. Methods: 134 mice were randomly divided into 3 groups: normal control group (nong24) treated with PQ (nd55, actual survival 24) and DHA treated group (nong55, 35 survived).
