α-半乳糖神經(jīng)酰胺對小鼠肺iNKT細(xì)胞數(shù)量、活性和氣道炎癥反應(yīng)的影響

【摘要】:目的觀察α-半乳糖神經(jīng)酰胺(α-GalCer)對小鼠肺iNKT細(xì)胞數(shù)量、活性和氣道炎癥反應(yīng)的影響。方法 12只野生型Balb/c小鼠隨機(jī)分為α-GalCer治療組和正常對照組,每組6只;12只CD1d~(-/-)Balb/c小鼠隨機(jī)分為CD1d~(-/-)α-GalCer治療組和CD1d~(-/-)對照組,每組6只。野生型和CD1d~(-/-)α-GalCer治療組小鼠單次腹腔注射α-GalCer 2μg,并用等量PBS對照。分別采用流式細(xì)胞儀檢測小鼠肺iNKT細(xì)胞、IL-4~+和IFN-γ~+i NKT細(xì)胞的數(shù)量;HE和PAS染色觀察肺組織炎癥和氣道杯狀細(xì)胞增生;姬姆薩染色檢測肺泡灌洗液(BALF)細(xì)胞總數(shù)和分類計(jì)數(shù);ELISA法檢測BALF中IL-4、IL-5、IL-10、IL-13水平和脾細(xì)胞培養(yǎng)上清液中IL-4、IL-5、IL-13水平。結(jié)果野生型α-GalCer治療組小鼠肺iNKT細(xì)胞、IL-4~+和IFN-γ~+i NKT細(xì)胞的數(shù)量明顯高于野生型正常對照組(P0.05)。野生型α-GalCer治療組小鼠肺組織無炎癥改變和基底膜杯狀細(xì)胞增生;BALF中細(xì)胞總數(shù)和分類計(jì)數(shù)、BALF及脾細(xì)胞培養(yǎng)上清液中IL-4、IL-5和IL-13水平與野生型對照組比較差異無統(tǒng)計(jì)學(xué)意義(P0.05)。野生型α-GalCer治療組小鼠BALF中IL-10水平明顯高于野生型正常對照組(P0.01),但CD1d~(-/-)α-GalCer治療組小鼠BALF中IL-10水平與野生型正常對照組和CD1d~(-/-)對照組比較差異無統(tǒng)計(jì)學(xué)意義(均P0.05)。結(jié)論腹腔注射α-GalCer可以增加小鼠肺iNKT細(xì)胞數(shù)量和活性,但不能誘導(dǎo)氣道炎癥反應(yīng)。
[Abstract]:Objective to investigate the effects of 偽 -galactose-ceramide (偽 -GalCer) on the number, activity and airway inflammation of iNKT cells in mice. Methods Twelve wild-type Balb / c mice were randomly divided into 偽 -GalCer treatment group and normal control group. Six mice in each group were randomly divided into two groups: CD1d- / (-r-) 偽 -GalCer treatment group and CD1d- (-r-) control group with 6 mice in each group. Mice in wild-type and CD1d- 偽 -GalCer group were given a single intraperitoneal injection of 偽 -GalCer 2 渭 g, and the same dose of PBS was used as control. The number of IL-4 ~ and IFN- 緯 ~ I NKT cells were detected by flow cytometry. The inflammation and goblet cell proliferation of the airway were observed by HE and pas staining. The total number of alveolar lavage fluid (BALF) cells and their classification count were detected by Giemsa staining. The levels of IL-4, IL-5, IL-10 and IL-13 in BALF and spleen cell culture supernatant were detected by Elisa. Results the number of IL-4 ~ and IFN- 緯 ~ I NKT cells in the wild-type 偽 -GalCer treated group was significantly higher than that in the wild-type normal control group (P0.05). There were no inflammatory changes in lung tissue and the total number and classification of BALF cells in the wild-type 偽 -GalCer treated mice. There was no significant difference in the levels of IL-4 IL-5 and IL-13 in the supernatant of spleen cell culture between the wild-type 偽 -GalCer group and the wild-type control group (P0.05). The level of IL-10 in BALF of wild-type 偽 -GalCer treated mice was significantly higher than that of wild-type normal control group (P0.01), but the level of IL-10 in BALF of wild-type 偽 -GalCer treated group was not significantly different from that of wild-type normal control group and CD1d- / (-) control group (P0.05). Conclusion Intraperitoneal injection of 偽 -GalCer can increase the number and activity of iNKT cells, but can not induce airway inflammation.
