【摘要】：目的:建立銅綠假單胞菌(Pseudomonas aeruginosa,PA)慢性肺部感染合并免疫低下大鼠模型,為相關(guān)模型建立與研究提供依據(jù)。方法:將實(shí)驗(yàn)大鼠分為空白對照組(A組)、肺部感染組(B組)、免疫低下組(C組)、肺部感染合并免疫低下組(D組)。以40 mg·kg~(-1)的劑量給予C、D2組大鼠腹腔注射環(huán)磷酰胺(cyclophosphamide,CTX),連續(xù)給藥3 d,造模成功后以相同劑量每周注射一次;運(yùn)用氣管插管法,構(gòu)建B、D組大鼠慢性肺部感染。在模型建立期間,以固定時(shí)間對大鼠肺載菌量、肺部病理學(xué)變化、免疫學(xué)指標(biāo)變化、T細(xì)胞亞群等指標(biāo)進(jìn)行考察。結(jié)果:HE染色結(jié)果顯示,B、D組大鼠在造模成功后肺部產(chǎn)生病理學(xué)病變,并有較明顯的中性粒細(xì)胞聚集等現(xiàn)象。與A組相比,B組與D組大鼠外周血中性粒細(xì)胞有不同程度的增高,C組與D組白細(xì)胞數(shù)目有所下降,實(shí)驗(yàn)組大鼠CD_4~+/CD_8~+有顯著增高。結(jié)論:通過該實(shí)驗(yàn)方法,成功建立PA慢性肺部感染合并免疫低下大鼠模型,成功率高、死亡率低,適于采取該方法進(jìn)行相關(guān)研究。
[Abstract]:Objective: to establish a rat model of chronic pulmonary infection of Pseudomonas aeruginosa PA with hypoimmunity, and to provide evidence for the establishment and study of the model. Methods: the experimental rats were divided into blank control group (group A), pulmonary infection group (group B), hypoimmunity group (group C) and pulmonary infection combined with hypoimmunity group (group D). The rats in the 40 mg kg ~ (-1) group were given cyclophosphamide CTX intraperitoneally for 3 days, and then the rats were injected with the same dose once a week after the successful establishment of the model, and the chronic pulmonary infection was constructed by tracheal intubation. During the establishment of the model, fixed time was used to investigate the changes of lung bacterial load, lung pathology, immunological indexes and T cell subsets in rats. Results the pathological changes of lung and neutrophil aggregation were found in the lung of rats in group B and D after the successful establishment of the model. Compared with group A, neutrophils in peripheral blood of rats in group B and group D increased to some extent, and the number of leukocytes in group C and group D decreased, and CD4 ~ / CD8 ~ increased significantly in experimental group. Conclusion: the model of PA chronic pulmonary infection combined with hypoimmunity was successfully established by this method, with high success rate and low mortality rate.
