茯苓、薏苡仁、冬瓜仁對肺纖維化大鼠肺組織TGF-β1、TNF-α影響的實驗研究

【摘要】:目的:借助大鼠肺纖維化動物模型,通過檢測大鼠肺組織中TGF-β1、TNF-α的表達水平,探討茯苓、薏苡仁、冬瓜仁治療肺纖維化的療效機制,為茯苓、薏苡仁、冬瓜仁的臨床應(yīng)用提供實驗依據(jù)。 方法:清潔級健康雄性Wistar大鼠,隨機分為6組,每組12只,除正常對照組外,模型對照組、醋酸潑尼松組、茯苓組、薏苡仁組、冬瓜仁組以支氣管內(nèi)注射博萊霉素的方法制作肺纖維化動物模型,于造模后第2天給予相應(yīng)藥物干預(yù);觀察大鼠一般情況,給藥后14天處死6只大鼠,28天處死余下大鼠,采集動物肺組織,光鏡下觀察大鼠肺組織病理變化情況,免疫組化法檢測大鼠肺組織中TGF-β1、TNF-α的表達情況。采用SPSS17.0統(tǒng)計軟件統(tǒng)計學(xué)處理相關(guān)實驗數(shù)據(jù)。 結(jié)果:實驗第26天模型對照組死亡1只大鼠,各組大鼠存活率比較均無顯著差異(P0.05)。博萊霉素干預(yù)的大鼠出現(xiàn)體重增長緩慢,甚至呼吸困難的癥狀。與正常對照組比較,模型對照組大鼠肺組織出現(xiàn)明顯的肺纖維化,且TGF-β1、TNF-α表達顯著高于正常對照組(P0.05),提示動物模型復(fù)制成功。與模型對照組比較,各治療組明顯改善肺纖維化模型大鼠肺組織的病理學(xué)改變,且TGF-β1、TNF-α水平均顯著低于模型對照組(P0.05);醋酸潑尼松組TGF-β1、TNF-α水平均低于其他各治療組(P0.05);茯苓組TGF-β1水平在14天、28天均低于薏苡仁組、冬瓜仁組(P0.05),薏苡仁組、冬瓜仁組之間比較無顯著性差異;茯苓組TNF-α水平在28天低于薏苡仁組、冬瓜仁組(P0.05),TNF-α水平在14天三組之間無顯著性差異。綜上所述,三個中藥組均可改善指標,茯苓組優(yōu)于薏苡仁組、冬瓜仁組,但均不及醋酸潑尼松組改善程度顯著。 結(jié)論:茯苓、薏苡仁、冬瓜仁均能有效緩解肺纖維化大鼠的癥狀表現(xiàn),抑制肺組織中TGF-β1及TNF-α的表達,推測三藥可通過抑制TGF-β1及TNF-α的表達來延緩博萊霉素誘發(fā)的肺纖維化進程。
[Abstract]:Objective: To explore the therapeutic mechanism of Poria, coix seed and winter melon kernel in the treatment of pulmonary fibrosis by using rat model of pulmonary fibrosis and the expression level of TGF- beta 1 and TNF- alpha in rat lung tissue, and provide experimental basis for the clinical application of Poria cocos, coix seed and winter melon kernel.
Methods: healthy male Wistar rats were randomly divided into 6 groups, with 12 rats in each group. In addition to the normal control group, the model control group, the prednisone acetate group, the Poria group, the Coix seed group, the winter melon kernel group were injected with bleomycin in the bronchus to make the pulmonary fibrosis animal model, and the corresponding drug intervention was given second days after the model. 14 days after administration, 6 rats were killed and the remaining rats were killed for 28 days. The lung tissue was collected. The pathological changes of lung tissue were observed under light microscope. The expression of TGF- beta 1 and TNF- a in the lung tissue of rats was detected by immunohistochemical method. The data of the related experimental data were used by the statistical software of SPSS17.0.
Results: 1 rats were killed in the twenty-sixth day model control group. There was no significant difference in the survival rate of the rats in each group (P0.05). The rats with bleomycin intervened the slow weight growth and even the symptoms of dyspnea. Compared with the normal control group, the lung tissue of the model control group had obvious pulmonary fibrosis, and the expression of TGF- beta 1 and TNF- a was significant. Compared with the model control group, the pathological changes of lung tissue in the pulmonary fibrosis model rats were obviously improved, and the level of TGF- beta 1 and TNF- a was significantly lower than that of the model control group (P0.05), and the TGF- beta 1 and TNF- alpha water of the prednisone acetate group were lower than those of the other treatment groups (P0.05). The level of TGF- beta 1 in Poria group was lower than coix seed group in 14 days and 28 days, but there was no significant difference between winter melon kernel group (P0.05), coix seed group and winter melon kernel group. The level of TNF- alpha in Poria group was lower than coix seed group, winter melon kernel group (P0.05), and TNF- alpha level between the three groups in 14 days. All of the three Chinese medicine groups could improve. The index, Fuling group was better than that of Coix seed group, semen Benincasae group, but were less than the prednisone group improved significantly.
Conclusion: tuckahoe, coix seed and winter melon kernel can effectively alleviate the symptoms of pulmonary fibrosis in rats and inhibit the expression of TGF- beta 1 and TNF- alpha in lung tissue. Three drugs can delay the progression of bleomycin induced pulmonary fibrosis by inhibiting the expression of TGF- beta 1 and TNF- alpha.
